摘 要:目的探讨沙利度胺(THD)联合替莫唑胺(TMZ)对人胶质母细胞瘤细胞株LN229黏附、侵袭能力的影响及其机制。方法LN229分别用加入DMSO(对照组)、THD(THD组)、TMZ(TMZ组)及THD+TMZ(联合组)的培养液培养,观察细胞的贴壁过程及形态变化。用流式细胞仪检测各组LN229中整合素αvβ3的表达。用黏附抑制实验与细胞侵袭实验测定细胞黏附及侵袭性。结果对照组和TMZ组LN229迅速形成伪足并与其他细胞接触形成单细胞层,整合素αvβ3均有较高表达。THD组和联合组LN229的伪足变短,形态有变圆倾向,整合素αvβ3表达减弱,其中贴壁细胞整合素αvβ3的阳性表达率分别为对照组的81.8%与77.9%,平均荧光强度(MFI)分别为对照组的66.8%、53.7%;悬浮细胞整合素αvβ3阳性表达率分别为对照组的63.7%、55.0%,MFI分别为对照组的49.8%、54.1%。THD组、TMZ组及联合组平均LN229黏附抑制率分别为25.23%、0.88%、26.83%。THD组及联合组平均每视野侵袭细胞数目较对照组和TMZ组减少(P均<0.05)。结论THD对LN229具有黏附抑制作用,对其黏附、迁移过程的抑制与细胞整合素αvβ3的表达有关。THD与TMZ间无协同作用。Objective To explore the inhibition effect and mechnaism of the combination with thalidomide(THD) and temozolomide(TMZ) on adhesion, invasion and integrin expression in human glioblastoma cell llne LN229. Methods LN229 cells were cultured in DMED medium supplanted with DMSO ( control group), THD ( THD group ), TMZ ( TMZ group) and THD + TMZ( combine group) ,and been observed with inverted microscope. Flow cytometry was used to analyze the integrins expression of LN229. The effect of THD and TMZ on the migration and invasion ability of LN229 was meatured by scratch model and transwell chamber invasion models. Results LN229 ceils in control group and TMZ group formed filopodia and adhered to other cells to form a cell-monolayer. Integrin αvβ3 intensively expressed in both two groups. The cells treated with THD and THD + TMZ tended to maintain a round shape by contracting the fdopodia. The integrin αvβ3 attached positive cells in THD and combine groups decreased to 81.8% and 77.9% of those in the control group, the mean fluorescence intensities (MFI) decreased to 66.8% and 53.7% ;the integrin αvβ3 positive suspended cells in THD and combine groups decreased to 63.7% and 55.0% of those in the control group,the MFI decreased to 49.8% and 54.1%. The adhesion-inhibiting late of THD,TMZ and combine groups was 25.23% ,0.88% and 26.83%. There were less invaded cells in THD and combine groups than that in the other two groups ( P 〈 0.05 ). Conclusions THD inhibits LN229 adhension and invasion. The inhibition is related to decrease of integrin αvβ3, but there's not synergistic effect in combination with TMZ.
