摘 要:通过利用非互补的PCR-RFLP技术检测了12个鸡群体378个样本在Mx基因编码序列第1892位的等位基因.结果表明:鸡Mx基因经PCR-RFLP扩增,获得长度约为100 bp的基因片段,在1892位发生A→G的突变;鸡Mx的抗病性(A)和易感性(G)等位基因频率分别为38%和62%,且分布于西南亚的BAN、PKD、PMA和Pal 4个鸡群体中的抗病基因频率明显高于其他群体.A G/A SNP at nucleotide position 1,892 of coding sequence of chicken Mx gene (a single amino acid substitution between Asn and Ser at position 631 in the chicken Mx protein) has been previously demonstrated to confer susceptibility/resistance to avian viral diseases. In this study,it was determined the resistant or susceptible Mx allelic gene in 378 samples from 12 chicken populations by using the mismatched PCR-restriction fragment length polymorphism (RFLP) technique. The rates of the resistant Mx gene and susceptible gene were 38 % and 62 %, respectively. And it is very interesting that the frequency of the resistant Mx gene in 4 populations from Southwest Asia (BAN, PKD, PMA and PAL) was obviously higher than the others.
