H460和H446肺癌细胞系Cyclin E表达及其临床意义的研究

摘  要:目的:检测细胞周期蛋白E(Cyclin E)在人肺癌细胞系中的表达水平,探讨Cyclin E与人肺癌发生及其生物学行为的关系。方法:采用双重胸苷阻断法分别对人胚肺正常细胞系MRC-5和大细胞肺癌细胞系NCI-H460及小细胞肺癌细胞系NCI-H446进行同步化处理,流式细胞仪检测其同步化效果;应用蛋白质印迹法检测Cyclin E在上述3种同步化处理细胞中的表达水平。结果:Cyclin E蛋白在肺癌细胞系NCI-H460和NCI-H446中的表达水平明显高于人胚肺正常细胞系MRC-5,差异有统计学意义,P<0.01。同时比较两种肺癌细胞系,NCI-H460细胞的CyclinE蛋白的表达水平明显高于NCI-H446细胞,P<0.01。结论:CyclinE蛋白与肺癌的发生及生物学行为可能具有相关性。OBJECTIVE: To detect the expression of Cyclin E in human lung cancer cell lines, and study the relationship between Cyclin E and the development and biological behavior of lung cancer. METHODS: The normal human lung cell line MRC 5, the large-cell lung cancer cell line NCI-H460 and small cell lung cancer cell line NCI-H446 were synchronized by the method of double thymidine block. And the results were analyzed by flow cytometry, then the expression of Cyclin E in three cell lines were detected with Western blot. RESULTS: The expression level of Cyclin E protein in NCI-H460 and NCI-H446 was significantly higher than that in MRC-5(P〈0.01). At the same time, comparing the two lung tumor cell lines, the expression level of Cyclin E protein in NCI-H460 was dramatically higher than that in NCI-H446(P〈0.01). CONCLUSION: Cyclin E protein may play an role in the development and biological behavior of lung cancer.
