乳腺癌组织OPN和p21ras表达及其与预后相关性的研究

摘　　要：目的:探讨骨桥蛋白(OPN)和p21ras在乳腺癌组织中的表达及意义。方法:采用SP法检测96例乳腺癌组织中OPN和p21ras的表达情况,分析其表达与5年生存率的关系。结果:在96例乳腺癌组织中,OPN阳性表达率为54.2%,在Ⅰ、Ⅱ、Ⅲ级癌组织中的阳性率分别为38.1%、52.4%和74.1%,在有、无淋巴结转移组阳性率分别为70.7%和41.8%,在生存期≥5年和<5年的表达率分别为39.1%和84.4%;p21ras阳性表达率为43.8%,在Ⅰ、Ⅱ、Ⅲ级中癌组织中阳性率分别是33.3%、45.2%和51.9%,在有、无淋巴结转移组阳性率分别为51.2%和38.2%,在生存期≥5年和<5年的表达率分别为32.8%和65.6%。OPN和p21ras在乳腺癌组织中的表达呈显著的正相关,r=0.52,P=0.00。OPN的表达与5年生存率呈明显负相关,r=-0.34,P=0.001。p21ras的表达与5年生存率呈明显负相关,r=-0.319,P=0.002。结论:OPN与p21ras的表达与乳腺癌的生物学行为有关,两者存在一定的协同性,联合检测可能更有助于判断乳腺癌患者的预后。OBJECTIVE： To investigate the expressions of osteopontin （OPN） and p21ras and significance in breast carcinoma. METHODS： SP imrnunohistochemical technique was employed to determine the expressions of OPN and p21ras in 96 breast carcinomas and analyze their relationship with the survival of 5 years. RESULTS： The positive rate of OPN was 54. 2%. The postive rates of it were 38. 1%, 52. 4%, 74. 1% in carcinomas with grade Ⅰ, Ⅱ and Ⅲ, respectively, and 70. 7% and 41.8% in the lymph nodes positive group and negative group, 39.1% and 84.4% among patients whose survival time ≥5 years and 〈5 years respectively; the positive rate of p21ras was 43. 8%. The positive rates of it were 33. 3%, 45. 2%. 51.9% in carci nomas with grade Ⅰ, Ⅱ and Ⅲ respectively, and 51.2% and 38. 2% in the lymph nodes positive group and negative group, 32.8% and 65. 6% among patients who survived more than 5 years and patients who sur vived less than 5 years respectively. OPN and p21ras were significantly correlated（r=0. 52,P=0. 00）. The expression of OPN had a significantly negative correlation with the survival of 5 years（r=-0. 34, P=0. 001）. The expression of p21ras had a significantly negative correlation with the survival of 5 years（r=-0. 319, P =0. 002）. CONCLUSIONS： Both OPN and P21 expressions are significantly associated with the bio logical behavior of breast carcinoma, and correlated each other. Co-examination for their expressions may help us evaluate the prognosis of the patients with breast carcinoma.
