摘 要:采用四氮唑蓝(MTT)分析检测10例骨肉瘤细胞对氨甲喋呤(MTX)、阿霉素(ADR)、顺铂(CDDP)、长春新碱(VCR)、氮芥(NM)、丝裂霉素(MMC)的药物敏感性。其样本的检测率达9O%。骨肉瘤细胞对化疗药物的敏感性不一,其50%细胞生存的药物浓度(IC50)差异很大。在临床用药剂量下,6种化疗药物的平均生存率之间差异有显著性(P<0.01),其中ADR除与CDDP之间差异无显著性外,与其它相比差异有显著性。同时观察到6种药物的剂量效应曲线也不相同。因而认为此结果不仅可作为一种药物敏感性试验的方法指导临床选择化疗药物,而且提示骨肉瘤细胞的药物作用模式。MTT assay was used to detect chemosensitivity to methotrexate (MTX), adriamycin (ADR), cisplatin (CDDP), vincristine (VCR) , nitrogen mustard (NM) and mitomycin (MMC) in 10 cases of osteosarcoma cells. The evaluability rate of all specimens is as high as 90 %. The sensitivity of osteosarco-ma cells to chemotherapeutic drugs is not always the same, its IC50 varies greatly. There is significant difference in the average survival rates of 6 kinds of chemotherapeutic drugs (P<0.01), and there is obvious difference between ADR and other drugs except CDDP. Their dose-effect curves are not the same. Therefore, we realized that the results we obtained could help to choose drugs in clincal practice as a method of chemosensitivity testing and reveal the model of drug effect on osteasarcoma cells.
