摘 要:目的探讨磁性纳米四氧化三铁颗粒(Fe3O4-MNPs)逆转人卵巢癌顺铂耐药细胞株(SKOV3/DDP)耐药性的可能性。方法将SKOV3/DDP细胞分为对照组、顺铂(DDP)组、Fe3O4-MNPs组和DDP+Fe3O4-MNPs组,体外培养后MTT法测定细胞生长半数抑制浓度(IC50),流式细胞术测定细胞凋亡及电感耦合等离子体原子发射光谱(ICP)法测定细胞内DDP含量。结果DDP组IC50为39.31μM,DDP+Fe3O4-MNPs组为17.4μM(P<0.05)。DDP+Fe3O4-MNPs组细胞凋亡率较DDP组明显提高(P<0.05)。DDP+Fe3O4-MNPs组细胞内顺铂含量明显高于DDP组(P<0.05)。结论Fe3O4-MNPs可与化疗药物顺铂协同,逆转卵巢癌细胞对DDP的耐受性。其作用可能是通过提高细胞内药物聚集量实现的。Objective To study the possibility of Fe3O4-MNPs reversing cisplatin (DDP) resistance to ovarian carcinoma cell (SKOV3/DDP). Methods The SKOV3/DDP strains were divided into DDP treatment (group A), Fe3O4 magnetic nanoparticles treatment (group B), DDP + Fe3 O4- MNPs treatment(group C), and control group(E). The proliferation inhibition (median inhibitory concentration, IC50 ) was measured by MTT, the apoptosis by flow cytometry, and the intracellular DDP level by inductively-coupled plasma atomic emission spectrometry (ICP). Results The ICs0 in group C was 17.4 μM, which was significantly less than 39.31 μM in group A (P〈0.05). The apoptosis rate of SKOV3/DDP cells was higher in group C than that in group A (P〈0.05). The intraeellular cisplatin level in group C was higher than that in group A (P〈0. 05). Conclusion Fe3 O4-MNPs can reverse the DDP resistance to the ovarian carcinoma cell (SKOV3/DDP), which may be related to its effect of increasing the intracellular accumulation of DDP.
