摘　　要：目的探讨C反应蛋白（CRP）对多发性骨髓瘤细胞株U266细胞株生长的影响及其机制。方法将CRP（浓度为0、5、10、20mg／L）与U266细胞株作用24h后，血液分析仪检测细胞增生情况；采用反转录-聚合酶链反应（RT—PCR）方法检测凋亡相关基因Survivin和HSP-90α的表达。结果培养24h后，U266细胞增生率明显提高（P〈0．05），且与CRP浓度成正比，CRP浓度为20mg／L时，细胞增生最明显；U266细胞表达Survivin、HSP-90α，其表达量随CRP浓度的增高而增高（P〈0．05）。结论CRP可促进U266细胞株增生，并通过调节凋亡抑制蛋白Survivin及HSP-90α表达的增高发挥其诱导U266细胞增生的作用。Objective In the present study, we examined the expression changes of Survivin/HSP90α, in C-reactive protein（CRP） treated human muhiple meyloma （MM） cell lies. Methods The human MM cell lines were cultured for 3 days and incubated with human CRP（0.5, 10, 20 mg/L for 24 hours） and analyzed by blood analyser for proliferation ratio. The effects of CRP in MM cell lines in various concentrations on survivin/HSP-90α mRNA expression were examined by RT-PCR. Results Proliferation ratio of U266 cell line was signitieantly increased, and the expression of survivin/HSP-90α were upregulated （gene promoting cell survival） at mRNA levels in proportion to increased CRP concentrations. Conclusion These results demonstrated that proliferation ratio, survivin and HSP-90α could be regulated by CRP in MM cells.
