摘 要:目的:通过检测胆管癌组织中多药耐药相关蛋白1(MRP1)和谷胱苷肽S转移酶-π(GST-π)的表达情况,探讨其耐药的临床意义。方法:收集21例手术切除的胆管癌标本作为实验组,另取8例胆管炎症标本作为对照组,采用免疫组织化学方法检测标本中MRP1及GST-π蛋白的表达,RT-PCR法检测标本中MRP1及GST-πmRNA的表达。结果:MRP1和GST-π蛋白在胆管癌组织中的表达阳性率分别80.1%和66.7%,显著高于对照组(P<0.05),MRP1和GST-πmRNA在胆管癌组织中的表达阳性率分别76.2%和66.7%,显著高于对照组(P<0.05),上述指标与患者性别、年龄、肿瘤病理分期、分化程度及淋巴结转移均无关(P>0.05)。结论:MRP1、GST-π在未经过化疗的胆管癌组织中均有不同程度的高表达;胆管癌的原发性多药耐药可能与MRP1、GST-π有关。Objective: To investigate the expression of multidrug resistance- related protein1 (MRP1), glutathione Stransferase-π(GST- π) in the patients with cholangiocarcinoma. Methods: The expressions of MRP1 and GST-π in cases of cholangiocarcinoma were measured by immunohistochemistry and RTPCR methods, while 8 cases of choledochitis served as the control group. Results: The positive rates of MRP1, GST-π protein in cholangiocarcinoma were 80.1% , 66.7% respectively, significantly higher than those of control group (P〈0.05). Their expressions were not related to gender, age, staging, differentiation and lymph node metastasis (P〉0.05). Conclusion: MRP1 and GST-π are overexpressed in various degrees in chotangiocarcinoma without chemotherapy, and they may be related to the primary multidrug resistance of the tumors.
