摘 要:目的探讨辣椒辣素诱导建立感觉神经损伤性盐敏感性高血压大鼠模型。方法新生雄性Wistar大鼠30只,在第1、2天分别皮下注射辣椒辣素50mg/kg,分2组给药。A组:辣椒辣素浓度小(2.5mg/ml),给药体积大(20μl/g);B组:辣椒辣素浓度大(10mg/ml),给药体积小(5μl/g)。每组15只,哺乳期(3周龄)后给予高盐(含4%NaCl)饮食1个月,然后用尾套法测血压。结果3周龄时,A组的成活率为46.7%,B组的成活率为86.7%。实验结束时,血压均在145~170mmHg。结论模型建立成功的关键:①辣椒辣素的浓度和新生大鼠的给药体积要合适;②皮下注射应严格无菌操作;③注意复温;④防感染。Objective To explore the experiences with establishment of salt-sensitive hypertension rat model induced by sensory denervation. Methods Thirty newborn male Wistar rats were given 50 mg/kg capsaicin subcutaneously on the 1st and 2nd day of life. The rats were then divided into two groups with 15 rats each. The group A received capsaicin at a small concentration (2.5 mg/ml) but large dosage volume(20 μl/g), while the group B received large concentration (10 mg/ml) but small dosage volume(5μl/g). Immediately after the weaning (3 weeks of life), the rats were fed on high sodium diet(4 % )for one month, then tail-cuff systolic blood pressure were measured. Results At three weeks of life, 46.7% of the group A rats were alive compared with 86.7% of the group B rats. At the end of study, the blood pressure of surviving rats measured 145-170 mm Hg. Conclusion The key to successful establishment of salt-sensitive hypertension model induced by sensory denervation may be determined by: ①appropriate concentration of capsaicin and dosage volume given to newborn rats; ③sterile manipulation during subcutaneous injec-tion;② proper rewarming;④prevention of infections.
