摘 要:目的利用细粒棘球绦虫微卫星序列为分型标记,建立高效、快速、简便的鉴定细粒棘球绦虫基因型技术。方法采用FAM和HEX两种不同荧光染料分别标记细粒棘球绦虫的Sca、Emsk、C106微卫星序列引物,利用荧光PCR-基因扫描技术鉴定新疆不同地区CE病人细粒棘球绦虫分离株的基因型。结果44例CE病人的66个分离株标本经PCR及微卫星标记鉴定均为细粒棘球绦虫,其中43例病人的65个细粒棘球绦虫分离株标本为G1基因型;1例病人的1个细粒棘球绦虫分离株标本为G6基因型。结论微卫星标记能够从基因水平快速、精确地鉴定新疆细粒棘球绦虫分离株的基因型,对细粒棘球绦虫的流行病学和致病性研究有重要价值。To set up a new method to verify the typical strains of Echinococcus granulosus and clarify the geographical distribution of its genotypes in Xinjiang, the Sca. Emsk and C106 microstellite sequences of E. granulosus were used as the typing markers and the genotypes of E. granulosus isolated from patients with echinocoecosis in various districts of Xinjiang were identified. It was found that 66 strains of E. granulosus were isolated from 44 patients with Echinococcosis ,in which 65 isolates from 43 patients were identified as G1 genotype and one isolate from patients was identified as G6 genotype by using PCR and microstellite markers. It is evident that the microstellite DNA markers can be used as a highly efficient, rapid and simple method to identify the genotypes of E. granulosus on genetic level and shows its important value in the studies of genetic polymorphism, epidemiology and pathogenesis of E. granulosus infection.
