摘 要:目的:探讨富血小板血浆对大鼠牙周组织再生的作用。方法:人工构建36只雄性SD大鼠下颌左右侧第一磨牙颊侧牙周骨缺损模型(共72个);随机分为4组,分别在缺损内填入富血小板血浆、富血小板血浆-Bio-Oss复合物、Bio-Oss(空白组不加任何材料),最后在所有缺损上盖Bio-Gide再生膜。分别在术后第7,14,21天处死动物,取组织标本,行HE、Masson染色,观察牙周组织修复情况,并测量14、21 d时HE切片中各组新生骨面积。结果:14、21 d时牙周新生骨面积(mm2)分别为:富血小板血浆组0.158±0.010,0.355±0.043;富血小板血浆-Bio-Oss组0.103±0.006,0.145±0.020;Bio-Oss组:0.060±0.005,0.111±0.010;空白组0.103±0.011,0.243±0.036;富血小板血浆组效果明显优于其它3组(P<0.05)。结论:应用PRP能有效促进大鼠牙周组织缺损的再生。AIM:To investigate the effect of plateletrich plasma (PRP) on the regeneration of rats periodontal tissue. METHODS : Mandibular first molars of 36 male SD rats were divided randomly into four groups, with eighteen teeth in each group. By surgery, periodontal fenestration defects were created at the buccal side of the molar roots. PRP was applied to the defects of the first group,while PRP/Bio - Oss to the second group and bio - Oss to the third group. The forth group was filled with nothing - as the control group. Finally, each defect was covered with a propotional piece of Bio - Gide membrane. The rats were euthanized on the 7th, 14th, and 21 st day after the surgery respectively, periodontal regeneration was observed by hematoxylin - eosin (HE) and Masson staining. The regenerated osseous tissue was calculated on the 14th and 21 st day by HE staining respectively. RESULTS:The areas of the regenerated osseous tissue (mm^2) on the 14th and 21st day were shown as follows: 0. 158 ± 0. 010, 0. 355 ±0. 043 (PRP),0. 103 ± 0.006,0. 145 ± 0.020 ( PRP/Bio - Oss ), 0.060 ±0. 005,0. 111 ± 0. 010 ( Bio - Oss ), and 0. 103 ± 0.011, 0.243 ±0.036 (control). Apparently the employment of PRP could produce the most desirable effect (P 〈 0.01 ). CONCLUSION : The application of PRP is the effective way to facilitate the regeneration of periodontal fenestration defects.
