大鼠局灶性脑缺血模型制备及取脑方法实验研究

摘　　要：目的:研究Wistar大鼠大脑中动脉脑缺血动物模型的规范制备方法和不同灌注方法灌注取脑的对照研究,总结关键环节和注意事项。方法:结扎颈总动脉(CCA)、颈外动脉(ECA),不结扎翼腭动脉,用适当粗细的鱼线从CCA盲插入颈内动脉(ICA)直到阻断大脑中动脉,拔出再灌注;规定时间4%多聚甲醛缓冲液心脏灌注取脑。结果:造模成功率高,造模动物临床表现典型;非灌注固定取脑,脑组织结构保留差;短期灌注固定取脑较好保留皮层和海马结构,但深部白质及核团结构保留差;长期灌注固定保留结构都好,但耗时长,固定液用量大,进行脑深部白质及核团结构研究效果好。结论:本模型制备方法易操作,注意事项详细,可信度高;研究皮层和海马缺血再灌注损伤选择短期灌注固定取脑尚可满足需要,如需研究深部白质及核团结构尚需长期灌注固定取脑。Objective： To explore a modified surgical approach to produce rat ischemia--reperfusion injury model and to study the perfusing fixative solution method of drawing the material, moreover, to summarize key points and critical items for attentions. Methods： After exposure and ligation of the common carotid artery （CCA） and the external carotid artery （ECA） ,the pterygopalatine artery were opened,the nylon string with its tip disposed was inserted into the internal carotid artery （ICA） from the common carotid artery. At defined time the rat was perfused with 4 % PFA buffer solution from heart , and then the rat brain was taken. Results： The rats developed typical symptoms and pathological manifestations after the surgery. Not perfusing fixative solution from the heart, the histological anatomy shape failed to remain well; short term perfusion of fixative solution kept better example cortex and hippocampus, but the white matter and the gray matter of the hidden poorly remained; long term perfusion could well keep them but took much time and used lots of solution, so a better effect could be achieved from the study on gray matter of the hiden. Coriclusion： This method is reliable and easy to operate, with rich critical items for attention. The method of drawing rat brain by short term perfusing fixative solution can be used to study the cortex and hippocampus＇ ischemical reperfusion injury. To study the gray matter of the hidden, it is necessary and acceptable to take long term perfusing.
