急性脑梗死血清MMPs水平及阿托伐他汀对其的影响

摘  要:目的检测急性脑梗死(acute cerebral infarct,ACI)患者治疗前后和对照组的血清基质金属蛋自酶-2(MMP-2)、基质金属蛋自酶-9(MMP-9)和组织型金属蛋白酶抑制剂-l(TIMP-1)的水平,评估基质金属蛋自酶在ACI的作用机制,探讨阿托伐他汀对ACI的治疗保护机制。方法实验分为ACI血脂正常组(A)、ACI高血脂组(AH组)、阿托伐他汀治疗急性ACI血脂正常组(AT组)、阿托伐他汀治疗ACI高血脂组(AHT组)。双抗体夹心ELISA法测定MMP-2、MMP-9和TIMP-1,生物化学法测定血脂。结果A、AH组经过阿托伐他汀治疗后MMP-2、MMP-9浓度明显降低,TIMP-1浓度明显增高(P<0.05)。结论阿托伐他汀能够降低急性脑梗死患者血清MMPs水平和增高血清TIMP-1的水平,阿托伐他汀可以调节细胞外基质动态平衡防治ACI。Objective To observe the influence of atorvastatin on serum thickniss of MMP-2, MMP-9 and TIMP-1 in acute cerebral infarct; to study its protection mechanisms. Methods In this study, the cases were divided into 4 groups: higher blood lipid level group without acute cerebral infarct (A), higher blood lipid level group with acute Cerebral infarct (AH), normal blood lipid level acute cerebral infarct group with atorvastatin treeatment (AT), higher blood lipid level acute cerebral infarct group with atorvastatin treeatment (AHT). MMP-2, MMP-9 and TIMP-1 were measured by enzyme linked immunosorbent assay; blood lipid level was measured by biochemistry. Results MMP-2, MMP-9 levels in A, AH ,group were significantly lower and TIMP-1 concentration significantly increased (P〈0.05) after atorvastatin treatment. Conclusion Atorvastatin can adjust the dynamic balance of extracellular matrix in the process of ACI.
