灯盏花素鼻腔喷雾剂对大鼠的鼻黏膜毒性研究

摘　　要：目的:评价灯盏花素鼻腔喷雾剂的鼻黏膜毒性。方法:采用大鼠模型法,分别于单次、多次给予灯盏花素鼻腔喷雾剂后,肉眼观察大鼠鼻腔给药侧鼻中隔黏膜;电镜观察制剂对鼻黏膜及纤毛形态、结构的影响。结果:大鼠给予灯盏花素鼻腔喷雾剂单次(1 d)、多次(7 d)后,其鼻中隔黏膜无充血及水肿现象,与空白对照组无显著性差异;在扫描电镜(SEM)下可见黏膜细胞表面纤毛密集、规则有序,未见纤毛断裂、倒伏等不正常现象;透射电镜(TEM)下所见纤毛细胞结构良好、纤毛内微管结构清晰、纤毛之间有微绒毛,细胞顶部胞质内线粒体等结构正常,与对照组基本相似。结论:灯盏花素鼻腔喷雾剂无明显鼻腔纤毛毒性。Objective： To investigate nasal toxicity of breviscapine nasal spray.Methods： Nasoseptal mucosa was observed after being treated with a single and multiple doses of breviscapine nasal spray in rats,respectively.The effect of spray on the appearance and structure of nasal mucosa and cilia was observed by electron microscope.Results： No congestion and edema phenomenon of nasal septum mucosa were found after a single（1 d） and multiple doses（7 d） of breviscapine nasal spray,compared with blank group.The surface cilia of mucosa cells were dense,regular and arranged in order,and no abnormal phenomenon of cilia fracture and lodging was obtained by SEM.The good structure of cilia cells,clear intra-micro-tubular structure cilia,microvilli were found in both cilia and normal structure of cellular mitochondria on the top of cells by TEM,compared with the control group.Conclusion： No significant nasal cilia toxicity of breviscapine nasal spray was found.
