摘 要:目的探讨银耳多糖(TP)对体外培养的心肌细胞及在体心肌的抗凋亡作用。方法体外细胞实验中,获取新生乳鼠心脏,消化分离心肌细胞并进行原代培养后,随机分为正常对照组、凋亡模型组和TP预处理组,于72h后行形态学研究及心肌细胞Na^+-K^+-ATP酶活力与细胞凋亡指数测定。动物实验中,选择清洁级ICR纯系小鼠100只,按体重随机分为5组:阴性对照组每天腹腔注射生理盐水,其余4组每天腹腔注射120mg/kgD-半乳糖以建立小鼠衰老模型,其中阳性对照组每天以生理盐水灌胃,3个TP干预组每天分别以100、200、400mg/kgTP灌胃。8周后处死小鼠,获取心肌标本进行组织学研究和凋亡指数、抗氧化指标检测。结果培养的各组乳鼠心肌细胞Na^+-K^+-ATP酶活力:正常对照组〉TP预处理组〉凋亡模型组(P〈0.05);心肌细胞凋亡指数:凋亡模型组〉TP预处理组〉正常对照组(P〈0.01)。动物实验中,与阴性对照组相比,阳性对照组及各TP干预组心肌均无明显病理学改变。TUNEL法检测心肌细胞凋亡指数显示:400mg/kgTP组〈200mg/kgTP组〈阳性对照组(P〈0.05)。心肌组织匀浆生化检测结果显示:200、400mg/kgTP组丙二醛和脂褐质含量低于阳性对照组(P〈0.05),且400mg/kgTP组低于200mg/kgTP组(P〈0.05);各TP干预组谷胱甘肽过氧化物酶和超氧化物歧化酶活性均高于阳性对照组(P〈0.05),其中400mg/kgTP组活性最高(P〈0.05)。结论TP能抑制氧化损伤诱导的体外培养心肌细胞凋亡的发生;对D-半乳糖致衰老模型小鼠心肌具有抗凋亡和抗氧化作用,且这种作用具有剂量相关性。Objective To observe the anti-apoptosis effects of Tremella Folysaccharides (TP) on cardio- myocytes in vitro and in vivo. Methods Cardiomyocytes were separated from neonate rats and were cultured in vitro. The cells were assigned to normal control group, apoptosis-induced group, and TP-pretreated group randomly. The morphology, Na^+ -K^+ -ATPase activity, and apoptosis index of cardiomyocytes were studied after 72 hours. In the intact animal study, 100 ICR mice were included and assigned randomly into 5 groups after paired by body weight: negative control group (peritoneal injected by saline daily) ; positive control group (peritoneal injected by D-galactose and intragastric administrated with saline daily) , and three TP-pretreated group groups (peritoneal injected by D-galactose and intragastric administered with 100, 200,400 mg/kg TP respectively). Mice were sacririced for histology, apoptosis index, and antioxidation studies after 8 weeks. Results Na^+ -K^+ -ATPase activity of cardiomyocytes were as follows: normal control group 〉 TP-pretreated group 〉 apoptosis-induced group ( P 〈 0.05 ) ; apoptosis index: apoptosis-induced group 〉 TP-pretreated group 〉 normal control group ( P 〈 0. 01 ). In the animal studies, there was no significant difference in myocardium pathology among groups. The apoptosis index detected by TUNEL was 400 mg/kg TP group 〈 200 mg/kg TP group 〈 positive control group ( P 〈 0. 05 ). Myocardium homogenate detection showed that malondialdehyde (MDA) and lipofuscin (LP) in 200 and 400 mg/kg TP groups were significantly lower than in positive control group ( P 〈 0. 05 ), MDA and LP in 400 mg/kg TP group was significantly lower than in 200 mg/kg TP group ( P 〈 0.05 ) ; and the activities of glutathione peroxidase (GSH-Px) and superoxide dismutase (SOD) in all TP-pretreating groups were significantly higher than in positive control group ( P 〈 0.05 ) ; among which the 400 mg/kg TP group had the highe
