摘 要:目的观察黄芪煎剂对大鼠的长期毒性反应,以评价其长期用药的安全性。方法80只Wistar大鼠随机分为高(180 g/kg)、中(90 g/kg)、低(45 g/kg)剂量组和对照组,给大鼠连续灌服黄芪煎剂90 d,给药期间每日观察动物的一般状况。停药后1,30 d分别处死动物,测量大鼠体质量,检测血、尿常规和血生化,测定主要脏器质量系数并做病理组织学检查。结果连续给药3个月对大鼠的一般状况,血、尿常规,主要脏器质量及其系数均无影响;心、肝、脾、肺、肾脏无病理变化。除停药1 d后高剂量组γ-GT高于对照组(P<0.05),其他各项生化指标与对照组比较均无显著性差异。高剂量组停药1 d后有8只大鼠胃黏膜呈现糜烂性损伤并散在点状出血点,光镜组织学检查为炎性改变。另2只胃黏膜正常。停药30 d后高剂量组有6只胃黏膜出现糜烂性损伤,但范围较小,程度较轻,且未见有出血点。结论黄芪长期用药对大鼠无明显毒性,可较大剂量长期服用。Objective It is to study the long term toxicity of Huangqi on rats and evaluate its safety of long term medication. Methods 80 Wistar rats were randomly divided into high dose (180 g/kg) group, middle dose (90 g/kg) group, low dose (45 g/kg) group and control group. The rats were consecutively drenched Huangqi for 90 days. The general status of the animals was observed daily in medication duration. The animals were killed at either 1 or 30 days after drug discontinuance. The body mass of the rats were measured. The routine control of blood and urine and the blood biochemistry were measured. The mass coefficient of major organ was determined and the histopathology detect was done. Results There were no influences of consecutively medication for three months on the general status, the routine control of blood and urine, the mass and coefficient of major organ of the rats. There were no pathological changes on heart, liver, spleen, lung and kidney. There were no significant differences on the biochemical indicators between high dose group and control group except γ- GT in high dose group at 1 d after drug discontinuance was higher than that in control group (P〈0.05). The gastric mucosa of eight rats showed erosion injury and sporadic punctiform petechia in high dose group at 30 days after drug discontinuance and the light microscope histology detection was inflammatory change. The gastric mucosa of the other two rats was normal in high dose group. The gastric mucosa of six rats showed erosion injury in high dose group at 30 days after drug discontinuance, but the extent was smaller, the degree was lighter and the petechia was not found. Conclusion Long term medication of Huangqi has no obvious toxicity on rats, so higher dose of Huangqi can be used for long term.
