摘 要:【目的】探索不同辐射抗拒鼻咽癌细胞CNE-2R和CNE-2细胞周期和形态学差异。【方法】细胞流式术检测细胞株CNE-2R和CNE-2在同步化前、同步化后及去同步化后24 h的细胞周期分布;透射电子显微镜观察CNE-2R和CNE-2形态学差异。【结果】撤去血清(细胞同步化)后24 h,CNE-2R和CNE-2中分别52.9%和52.1%细胞停滞于G0/G1期;加入血清(去同步化)24 h后发现,CNE-2细胞周期中G1期细胞下降到48.5%,而G2期细胞有所上升,但CNE-2R细胞中G1期细胞与同步化前相比反而增加(65.3%)。与CNE-2细胞(26.5%)相比,CNE-2R处于G2期的细胞比例(8.4%)则明显减少;CNE-2R细胞S期的比例(26.3%)与CNE-2细胞(25.1%)相仿;与CNE-2细胞相比,CNE-2R线粒体数量增多、体积增大、嵴清晰;内质网扩张,呈现板层状态;可见CNE-2R细胞内微腔的形成。【结论】鼻咽癌细胞的不同辐射抗拒机制与细胞周期分布和细胞器的形态学改变有关。[Objective] To detect cell cycle proportion distribution and observe morphology discrepancy in different radioresistant nasopharyngeal carcinoma (NPC) cells CNE-2R and CNE-2. [Methods] Cycle proportion distribution were detected by flow cytometry before synchronization, after synchronization, and 24 h after loss of synchronization and the morphology was observed with transmission electron microscope. [Results] After serum starvation for 24 h, there were 52.9% of CNE-2R and 52.1% of CNE-2 cell in G0/G1 stage respectively. However, the proportion of G1 cells in CNE-2 decrease to 48.5% but CNE-2R increase to 65.3% after serum re-stimulation 24 h. Compared to CNE-2 cells, the proportion of G2 cells in CNE-2R (8.4%) was lower than that in CNE-2 (26.5%); the proportion of S cells in CNE-2R and CNE-2 were similar (26.3% versus 25.1%). Compared with CNE-2 cells, CNE-2R had lager number and volume of mitochondria and contained expanded lamellar-formed endoplasmic reticulum and microcavities as well. [ Conclusion ] The discrepancy of cycle proportion distribution and morphology are present in different radioresistant NPC cell lines and related to radioresistance.
