摘  要:目的:研究重组人白细胞介素24体外对淋巴瘤raji细胞的生长抑制作用及其分子机制。方法:用rhIL-24蛋白作用于淋巴瘤raji细胞,MTT法检测rhIL-24蛋白对raji细胞的生长抑制作用,激光扫描共聚焦显微镜(LSCM)观察rhIL-24蛋白诱导raji细胞凋亡的形态学改变,流式细胞仪(FACS)检测rhIL-24蛋白诱导raji细胞的凋亡率及细胞周期改变,免疫细胞化学分析凋亡相关因子Bcl-2和Bax的改变。结果:rhIL-24蛋白作用于淋巴瘤raji细胞后,可以显著抑制raji细胞的生长,诱导G2/M期阻滞和凋亡,rhIL-24蛋白能使raji细胞中bcl-2的表达下调和bax的表达上调。结论:rhIL-24蛋白对淋巴瘤raji细胞具有显著的生长抑制及凋亡诱导作用,其作用机制可能与下调bcl-2和上调bax的表达有关。Objective:To explore the growth inhibition effect and its molecular mechanism of raji cells by the recombinant human interleukin 24 protein in vitro. Methods:The rhIL -24 was used to react with lymphoma cells line rail. The anti -tumor efficacy of rhIL -24 on raji cells was evaluated by MTT, and cell apoptosis was detected by Hoechest 33258 staining and laser scanning eonfocal microscopy(LSCM). The cell cycle and distribution and apoptotic peak was evaluated by flow cytometry. Bcl - 2 and Bax were explored by means of immunocytochemistry. Results: rhIL - 24 could obviously inhibit proliferation and induce G2/M phase arrest and cell apoptosis of raji cells, rhIL - 24 can also down - regulate the expression of bcl - 2 and up - regulate the expression of bax. Conclusion: rhIL - 24 can obviously inhibit proliferation and induce cell apoptosis of raji cells, down - regulating the expression of bcl - 2 and up - regulating the expression of bax may he one of the molecular mechanisms involved in the anti - tumor effect.
