有机锗GeM_(10)对体外培养黑色素瘤细胞DNA合成的抑制作用

摘　　要：ＧｅＭ１０是一种抗癌生物碱——金属配位药物。动物试验发现ＧｅＭ１０对实验肿瘤Ｓ１８０肉瘤有抑制作用，能明显延长艾氏腹水癌药瘤小鼠的存活时间。本实验用体外细胞培养技术检测了ＧｅＭ１０对Ａ３７５黑色素瘤细胞３Ｈ－ＴｄＲ掺入的影响，结果发现：ＧｅＭ１０能抑制Ａ３７５黑色素瘤细胞ＤＮＡ合成（Ｐ＜０．０１），浓度为１８０μｇ／ｍｌ时抑制率最大，可达７３％（Ｐ＜０．０１）；作用时间２４ｈ，各浓度ＧｅＭ１０抑制效果最强。该实验结果提示。GeM_(10) is a new organic germanium Compound-alkalaidmetal complex.Our previous date showed that GeM10 could inhibit S 180 Solid tumor and Ehrilish ascites in mice.Although GeM_(10) Showed strong antitumor effect in Vivo,the mechanism of the antitumor effect is still unclear.In this paper,we studed the antitumor cytostasis by3TdR in corporation test using A375 melanoma cell line.the results showed that GeM_(10) could inhibit significantly the 3TDR in corporation of A375 melanoma cell,70% inhibitory rate could be obtained at the concentration ranging from 180% μg/ml to 300 μg/ml GeM_(10).The optional inhibitory dose was 180 μg/ml GeM_(10),and the peak inhibition was at 24 hours.these findings suggest that GeM10 can directly inhibit the tumor growth and may have promise as a antitumor drug.
