摘  要:目的探讨新型气体信号分子硫化氢(H2S)对吗啡依赖大鼠伏核、海马cAMP含量的影响。方法将30只SD大鼠,随机分为生理盐水组、吗啡依赖组和吗啡依赖+NaHS组(每组10只)。按剂量递增原则,给SD大鼠皮下注射吗啡建立吗啡依赖大鼠模型,每天注射吗啡前30min给予吗啡依赖+NaHS组大鼠腹腔注射NaHS,其余各组注射等量的生理盐水。模型建立成功后化学分光光度法测定血浆H2S含量,放射免疫法检测伏核和海马cAMP的含量。结果依赖组大鼠血浆中H2S含量低于生理盐水组(P<0.05),伏核和海马cAMP含量均高于生理盐水组(P<0.05);吗啡依赖+NaHS组大鼠血浆中H2S含量高于吗啡依赖组(P<0.05),伏核和海马cAMP含量均低于吗啡依赖组(P<0.05),而与生理盐水组相近(P>0.05)。结论H2S可能引起吗啡依赖大鼠伏核、海马cAMP水平的变化。[Objective] To explore the effects of new gaseous signal molecule-hydrogen sulfide (H2S) to cAMP of the Aceumbens and the Hippoeampus levels in morphine-dependent rats. [Method] 30 SD rats were divided into normal saline, morphine-dependent and morphine-dependent+NariS group (10 each group). According to the princiele of increasing dose daily,morphine was injected by subcutaneous to establish morphine-dependent rat models, before morphine was injected 30 minutes daily, injected Naris to morphine-dependence + Naris rats, after morphine-dependent rat models were established, detected H2S content of plasma by spectrophotometry and the cAMP eontent of the aeeumbens and hippocampus by RIA (radioimmunoassay). [Result] H2S content of plasma decreased in dependent rats than the saline group (P 〈0.05), cAMP content in the Accumbens and Hippocampus were higher than normal saline group (P 〈0.05); H2S content of plasma increased in morphine-dependent + NariS rats than morphin-dependence group (P 〈0.05), cAMP level was lower in Aecumbens and Hippocampus than the morphine-dependent group (P 〈0.05), while similar to the saline group (P 〉0.05). [Conclusion] H2S may be involved in cAMP change of Aecumbens and Hippoeampus in morphine-dependent rats.
