摘 要:目的探讨外源性一氧化氮(nitric oxide,NO)对牙周膜细胞(periodontal ligament cells,PDLCs)增殖的抑制作用。方法培养人牙周膜细胞,将培养增殖的4~8代细胞分为两组,即实验组和对照组。实验组中加入不同浓度的硝普钠(sodium nitroprusside,SNP)作为NO的供体,对照组不加SNP。培养24h后用硝酸还原酶法检测细胞上清液中NO的含量;MTT检测活细胞数量,连续监测法检测碱性磷酸酶活性。结果24h后细胞上清液中NO含量随SNP浓度增高而增加;碱性磷酸酶活性随SNP浓度增加而降低;MTT实验显示经SNP作用后的的活细胞数明显较对照组少(P<0.05);去除SNP作用后牙周膜细胞仍能保持旺盛的增殖能力,且实验组不同浓度之间无明显差异(P>0.05)。结论NO对培养状态下PDLCs的增殖有抑制作用。Objective To investigate the inhibitive effect of nitric oxide (NO) on the proliferation of periodontal ligament cells (PDLCs) in human.Methods PDLCs were cultured,and 4-8th passage cultured PDLCs were used. The medium was supplemented with SNP as a donor of NO in vitro. The release of NO from SNP was measured by nitric acid deoxidize enzyme in the medium. The quantity of PDLCs proliferation was measured by MTT assay. The ALP activity of PDLCs were measured by continuous observation.Results The release of NO increased along with the enhancement of the SNP concentratio-n. However,the ALP activity decreased with the enhancement of the SNP concentration. The amount of living cells in experimental groups decreased compared with the control groups as shown by MTT test (P〈0.05). The PDLCs still had the proliferation ability when SNP was removed,and the concentration of SNP had no effect (P〉0.05).Conclusion Nitric oxide inhibits the proliferation of human PDLCs in vitro.
