摘 要:目的探讨结缔组织生长因子(connective tissue growth factor,CTGF)对人牙周膜成纤维细胞VEGF表达的影响.方法通过组织培养获得人牙周膜成纤维细胞(HPLFs);第5代HPLFs实验组细胞加入不同浓度CTGF(5、10、50、100ng/mL),对照组加入等量的蒸馏水,培养48h后,收集细胞爬片,免疫组化及计算机图像处理系统检测检测HPLFs中VEGF表达变化.结果通过组织培养获得细胞抗波形丝蛋白阳性,抗角蛋白阴性,为中胚层来源的成纤维细胞,符合HPLFs的形态学及免疫学特征.CTGF终浓度为5~100ng/mL,实验组VEGF表达强度高于空白对照组.CTGF终浓度为10ng/mL时VEGF表达强度增幅最大.结论CTGF干预可有效上调HPLFs中VEGF表达.Objective To study the effects of CTGF on VEGF expression in human periodontal ligament fibroblasts. Methods The HPLFs were obtained by tissue culture, and detected by immunohistochemical stain. CTGF (5, 10, 50, 100 ng/mL) were added into the HPLFs which were passed fifth generation, and cultured. 48 hours later, the cells were collected and analyzed by immunohistochemical stain and automatic image analysis system to detect the VEGF expression in treated HPLFs. Results HPLFs got from tissue culture were confirmed by morphology and immunohistochemical detection. The CTGF (5 - 100 ng/mL) up-regulated VEGF expression in HPLFs. The expression of VEGF reached the peak when the concentration of CTGF was 10 ng/mL. Conclusion CTGF can effectively increse the expresion of VEGF in HPLFs.
