摘 要:目的探讨乏氧显像剂18F-氟赤式硝基咪唑(18F-FETNIM)在食管癌细胞ECa109乏氧监测中的应用价值。方法传代至2~3代的ECa109细胞,常氧环境下分别予以0、2、4、6、8 Gy X线照射,加入等量18F-FETNIM显像剂后分别于5、30、60、120、180 min用γ井型测量仪测定其放射性计数,计算摄取率。结果18F-FETNIM在ECa109细胞中的摄取,0 Gy组细胞的摄取率随时间延长而增高,180 min达最高,其余各组细胞随照射剂量增加,摄取率峰值逐渐前移,且峰值均高于0 Gy组。结论18F-FETNIM可用于食管癌细胞乏氧情况监测,为临床优化化疗方案提供依据。objective To investigate the monitoring value of hypoxic tracer fluorine-18-fluoroerythronitroimidazole ( ^18 F- FETNIM) in esophageal cancer ECa109 cells line. Method ECa109 cell line, which had been passaged 2-3 times and cultured under normoxic environment, were given different doses of X-ray irradiation (0 Gy,2 Gy ,4 Gy ,6 Gy ,8 Gy), separately. The uptake of ^18F-FETNIM in cell line were counted at 5 min,30 min,60 m/n, 120 min, 180 min separately after adding the hypoxia imaging agent ^18F-FETN/M by Well γ-type measuring instrument. Result The cell line uptake rate of 0 Gy group increased continuously as the time pass and reached the peak at 180 rain ,while the other groups, the time to uptake peak were shortened and become shorter as the X-ray radiation dose enhanced step by step. All the peak uptake rate in X- ray radiation groups were higher than that in 0 Gy group. Conclusion The uptake of ^18F-FETNIM for esophageal cancer cells can be used for the monitoring of hypoxia, in order to optimize the clinical programs provide a basis for radiotherapy and chemotherapy.
