摘 要:目的研究重组腺病毒感染宫颈癌细胞Hela和乳腺癌细胞MCF-7的转染效率,并观察重组腺病毒介导的人肿瘤坏死因子(TNF)相关诱导凋亡配体(hTRAIL)基因对Hela、MCF-7生长的影响。方法将Hela和MCF-7细胞接种至12孔板,培育24h后加入不同感染滴度的重组腺病毒Ad-CMV-EGFP,用流式细胞仪检测其转染效率;将Hela细胞和MCF-7细胞接种至96孔板,24h后转染重组腺病毒Ad-CMV-hTRAIL,72h用四甲基偶氮唑盐(MTT)比色法检测肿瘤细胞的存活率。结果Ad-CMV-EGFP对Hela和MCF-7细胞的转染效率在一定范围内随着感染复数(MOI)的增加而增高,转染Ad-CMV-hTRAIL的Hela细胞的存活率明显低于转染Ad-CMV-EGFP的病毒对照组(P<0.05)。结论腺病毒对Hela和MCF-7细胞均有较高的转染效率,且腺病毒介导的hTRAIL基因对Hela细胞具有明显的体外抗瘤作用。Objective To study the infection efficiency of recombinant adenovirus to uterine cervix cancer cell and MCF-7, and observe the effect on the growth of uterine cervix cancer cell Hela and MCF-7 of hTRAIL gene mediated by recombinant adenovirus. Methods After Hela cells and MCF-7 cells were added to wells of 12-well plates and incubated for 24 h, the different MOI of Ad-CMV-EGFP virus were added into the 12-wells, infected by recombinant adenovirus, infection efficiency was detected by flow eytometry. The adenovirus( Ad- CMV-hTRAIL) were transfected after Hela cells and MCF-7 cells being added to the 96-well plates for 24 h, and the survival rates were detected using MTT method 72 h later. Results Within a certain range, the infection efficiency of recombinant adenovirus to uterine cervix cancer cell and MCF-7 increased with increasing of MOI. The survival rates of Hela cell transfected with Ad-CMV-hTRAIL were significant lower than the control group transfected with Ad-CMV-EGFP. Conclusions By recombinant adenovirus to infect uterine cervix cancer cell and MCF-7 can increase the infection efficiency, and hTRAIL gene mediated by adenovirus has anti-tumor effect on Hela in vitro.
