摘  要:背景:课题组以往的研究证实黄芪对体外培养的人表皮体外扩展生长和表皮干细胞增殖均具有促进作用,但它对人骨髓间充质干细胞的体外作用如何特别是是否具有诱导骨髓间充质干细胞向表皮样细胞分化的效应,仍不清楚。目的:观察黄芪体外定向诱导人骨髓间充质干细胞分化为表皮样细胞的可行性。设计、时间及地点:细胞学体外对比观察,于2006-09/2008-06在南昌大学第一附属医院完成。材料:骨髓来源于临床骨髓穿刺检查正常患者,无血液系统疾病和家族遗传病史。方法:用密度梯度离心结合贴壁法分离纯化培养人骨髓间充质干细胞,以含黄芪、胎牛血清、角质细胞无血清培养基组成的诱导培养液进行体外培养,以不含黄芪的上述培养液作为对照。主要观察指标:倒置显微镜下观察细胞形态学变化,免疫细胞化学染色法检测p63和广谱细胞角蛋白的表达。结果:经黄芪诱导后部分细胞由长梭形转变为扁园形,呈表皮样细胞形态特征;免疫细胞化学染色显示部分诱导细胞p63、广谱细胞角蛋白表达均呈阳性。结论:黄芪可诱导人骨髓间充质干细胞分化为表皮样细胞。BACKGROUND: Previous studies have verified that radix astragalis can promote human epidermis growth in vitro and epidermal stem cell proliferation. It is still unclear that its effects on human bone marrow mesenchymal stem cells (BMSCs) in vitro and that whether it can induce the differentiation of BMSCs into epidermis-like cells. OBJECTIVE: To investigate the feasibility of differentiation of human BMSCs into the epidermal-like cells induced by radix astragalis in vitro. DESIGN, TIME AND SETTING: The cytological in vitro controlled study was performed at the First Affiliated Hospital, Nanchang University from September 2006 to June 2008. MATERIALS: Bone marrow aspiration samples in this study were obtained sterilely from human donors without severe hematopietic and genetic diseases. METHODS: The human BMSCs were harvested, isolated and purified by combining density gradient centrifugation with adherent method. The cultured cells in induction group were treated with keratinocyte serum-free medium (KSFM) supplemented with radix astragalis and fetal bovine serum. No radix astragalis was added in control group. MAIN OUTCOME MEASURES: The morphological changes of the cells were observed with an inverted microscope. The expression of p63 transcription factor and cytokeratin was determined by immunocytochemical method. RESULTS: The some cells induced by radix astragalis showed the morphological changes from long fusiform-shaped to oblong shape. The expression of p63 transcription factor and cytokeratin was positive in these cells of induction group. CONCLUSION: BMSCs can differentiate into epidermal-like cells induced by radix astragalis in vitro.
