摘 要:目的:探讨大肠癌组织中Survivin及其剪接变异体Survivin-ΔEx3、Survi-vin-2B mRNA在大肠癌组织的表达规律。方法:采用逆转录-聚合酶链反应(RT-PCR)检测50例(高分化20例,中低分化30例)大肠癌组织标本中Survivin及其剪接变异体Survivin-ΔEx3、Survivin-2B mRNA表达。结果:Survivin、Survivin-ΔEx3及Survivin-2B mRNA在大肠癌中表达阳性率分别为70.0%、34.0%、26.0%,大肠癌高分化组和中低分化组Survivin mRNA阳性表达率有显著差异;Survivin-ΔEx3mRNA阳性表达率有上升趋势,Survivin-2B mRNA阳性表达率有下降趋势。50例大肠癌组织中表达Survivin-△Ex3和(或)Survivin-2B者均有Survivin表达。结论:提示Survivin在大肠癌的发展过程中具有重要作用;Survivin及其剪接变异体Survivin-ΔEx3具有抗凋亡作用;Survivin-2B促凋亡,具有保护作用;Survivin基因有望作为大肠癌生物治疗的一个良好分子靶点。Objective:To investigate the expression of Survivin and its splice variants Survivin-△Ex3,Survivin-2B mRNA in colorectal cancer. Methods:The expression levels of mRNA of Survivin and its splice variants Survivin-△Ex3 and Survivin-2B were in vestigated by reverse transcription polymerase chain reaction (RT-PCR) in 50 cases of colorectal cancer tissues (20 cases of high differentiation & 30 cases of moderate/poor differentiation).Results:The positive expression rate of mRNA of Survivin,Survivin-△Ex3 and Survivin-2B in colorectal cancer were 70.0%, 34.0% and 26.0% respectively. There were significant differences between the positive expression rates of mRNA of Survivin in high differentiation and moderation/poor differentiation colorectal cancer. The positive expression rates of mRNA of Survivin-△Ex3 had escalating trend.the positive expression rates of mRNA of Survivin-2B had moderating trend.The expression of Survivin have been found in any tissue where survinvin-△Ex3 and (or) Survivin-2B express.Conclusion:Survivin plays an important role in colorectal cancer. Together with its splice variants Survivin-△Ex3, Survivin is an inhibitor of apoptosis protein. Meanwhile, Survivin-2B is a promoting apoptosis factor. Survivin is an ideal molecular target for biotherapy of colorectal cancer.
