摘 要:以琼胶糖为原料,样品用干法灰化法处理后分别以超纯水和1.0mol/L盐酸溶解定容,利用离子色谱法和硫酸钡比浊法测定其硫酸基含量(以SO42-计)。结果表明,离子色谱法SO42-在1.0~15.0mg/L范围内线性关系良好(R2=0.9996),硫酸钡比浊法SO42-在6.0~30.0mg/L范围内线性关系良好(R2=0.9991);对同种琼胶糖样品进行6次重复测定,离子色谱法相对标准偏差为1.40%(n=6),具有较高的精密度,硫酸钡比浊法相对标准偏差为3.51%(n=6);对两种不同的琼胶糖样品分别进行两个水平的加标回收率实验,离子色谱法回收率为86.33%~97.40%,硫酸钡比浊法回收率为84.17%~96.67%。因此,离子色谱法和硫酸钡比浊法均能准确测定样品中硫酸基含量。Agarose was dry ashed and the ash was separately diluted with ultrapure water and 1.0 mol/L hydrochloric acid to volume for measuring sulfate group content by ion chromatography (IC) and barium sulfate turbidimetry (BST). The IC method exhibited a good linear relationship in the SO4^2- concentration range of 1.0-15.0 mg/L, with a correlation coefficient of 0.9996, the RSD of 6 repetitions was 1.40%, indicating a high precision, and the recoveries in at two spiked levels were from 86.33% to 97.40%; As for the BST method, the linear range was 6.0-30.0 mg/L and the correlation coefficient was 0.9991, the RSD of 6 repetitions was 3.51%, and the recoveries in at two spiked levels were from 84.17% to 96.67%.These results show that both IC and BST can provide an accurate determination of sulfate group in agarose.
