摘  要:目的:探讨失血性休克大鼠血清对人脐静脉内皮细胞内皮细胞蛋白C受体(EPCR)表达的影响。方法:采用酶联免疫吸附试验(ELISA)、逆转录聚合酶链反应(RT-PCR)、WESTERN BLOT技术分别对正常对照组、失血性休克(3h、12h、24h、72h)组大鼠血清培养的人脐静脉内皮细胞(HUVEC)可溶性EPCR(sEPCR)水平和EPCR mRNA及蛋白表达进行检测。结果:与正常对照组比较,休克3h、72h组sEPCR、EPCR的mRNA、蛋白含量均无明显变化(P均>0.05),休克12h、24h组sEPCR、EPCR的mRNA含量均显著增高(P均<0.05),EPCR的蛋白含量均显著降低(P均<0.05)。结论:失血性休克大鼠血清可以从基因、蛋白水平影响人脐静脉内皮细胞EPCR的表达。Objective:To investigate the effects of the serum in hemorrhagic shock rats and Shenfu injection on endothelial cell's thrombomodul and endothelial cell protein C receptor. Methods:The soluble endothelial protein C receptor (sEPCR) in supernatant , EPCR mRNA and protein of human umbilical vein endothelial cell were detected by enzyme linked Immunosorbent assay (ELISA), reverse transcription polymerase chain reaction (RT- PCR) , western bloting ( WB ) in endothelial cell culturing with serum, normal control, hemorrhagic shock (3h, 12h, 24h ,72h) group respectively. Results : The mean levels of sEPCR, the expression of EPCR mRNA and protein were no significant differences in hemorrhagic shock(3h,72h) group than that in normal control group ( all P〉0.05). The mean levels of sEPCR and the expression of EPCR mRNA were significantly higher in hemorrhagic shock(12h,24h) group than that in normal control group ( all P 〈 0.05 ), while the level of protein was lower than that in normal control group ( both P 〈 0.05 ). Conclusion: These data suggest that serum in hemorrhagic shock rats could effect sEPCR ,EPCR mRNA and protein of human umbilical vein endothelial cells expression.
