摘  要:目的:观察精子蛋白17(Sp17)的异常表达对体外培养的卵巢癌细胞顺铂和卡铂耐药性的影响。方法:将Sp17基因转染卵巢癌细胞系HO8910,采用流式细胞术测定目的蛋白的表达,获得稳定高表达Sp17的卵巢癌细胞系HO8910/Sp17。MTT法检测铂类药物对转染细胞HO8910/Sp17的生长抑制作用并与未转染的亲代细胞进行比较。结果:在有效剂量范围内,顺铂和卡铂对HO8910/Sp17细胞的抑制率明显低于亲本细胞HO8910,P<0.05。耐药指数顺铂1.4,卡铂1.38。结论:Sp17明显增加卵巢癌细胞对铂类化疗药物的耐药性。OBJECTIVE: To observe the effect of overexpres sion of human sperm protein 17(Sp17) on the chemoresistance of ovarian cancer ceils to cisplatin and earboplatin. METHODS: hSpl7 cDNA was transfected into human ovarian carcinoma cell line HO8910 to establish a cancer cell line with stable overexpression of Sp17. The expression of interest protein was examined by flow cy tornetry. The inhibitory effects of carboplatin and cisplatin on trans feted cancer cells HO8910/Sp17 and parent HO8910 were detected by the colorimetric MTT assay. RESULTS: The inhibition ratios of carboplatin and cisplatin on HO8910/Sp17 cells were significantly lower than those on parent cells (P〈0.05) at appropriate concentrations. The resistance indexes of cisplatin and carboplatin were 1.4 and 1.38, respectively. CONCLUSION: The overexpression of Sp17 can increase the ehemoresistance of ovarian cancer cells to platinum.
