摘 要:目的:研究胃泌素受体拮抗剂(丙谷胺)联合环氧合酶-2抑制剂(NS-398)对人大肠癌LoVo细胞增殖和细胞周期的影响。方法:人大肠癌LoVo细胞株培养于RPMI1640培养液中,加入丙谷胺6个稀释度1.0、3.0、5.0、6.0、8.0、10.0 mmol/L及NS-398 6个稀释度0.01、0.1、1.0、10.0、100.0、1000.0μmol/L连续培养24 h后用噻唑蓝(MTT)比色实验检测肿瘤细胞增殖抑制率;单用丙谷胺稀释度为6 mmol/L,单用NS-398稀释度为100.0μmol/L及两者的混和液作用24、48、72 h后检测肿瘤细胞抑制率;单用丙谷胺稀释度为6 mmol/L,单用NS-398稀释度为100.0μmol/L以及2者的混和液作用24 h后,用流式细胞仪测定细胞周期的变化。结果:丙谷胺与NS-398均能抑制LoVo细胞株增殖,丙谷胺与NS-398能协同作用,抑制大肠癌LoVo细胞增殖,且协同作用随药物作用时间的延长而增强。流式细胞仪检测表明两者均能抑制大肠癌LoVo细胞从G1期到S期的转化,两者联合用药抑制效果更为显著。结论:丙谷胺与NS-398均能抑制大肠癌LoVo细胞从G1期到S期的转化从而抑制LoVo细胞株的增殖,两者合用具有协同抗癌作用。Objective To explore effects of gastrin receptor antagonist proglumide and the specific cyclooxygenase (COX)-2 inhibitor NS-398 on the proliferation and the cell cycle of the large intestine carcinoma cell lines LoVo cultured in vitro. Methods Human colonic carcinoma cell lines LoVo were routinely cultured in RPMI 1640 medium supplemented with 10 % heat inactivated fetal calf serum. Subconfluent cell cultures were treated with proglumide of concentrations (1.0, 3.0, 5.0, 6.0, 8.0, 10.0 mmol/L) and NS-398 of concentrations (0.01, 0.1, 1.0, 10.0,100.0, 1000.0μmol/L) for 48 h. The growth and proliferation of LoVo ceils were analyzed with MTT assay. Subconfluent cell cultures were treated with proglumide at a concentration of 6.0 mmol/L with or without NS-398 at a concentration of 100.0 μmol/L for 24 h, 48 h and 72 h, then the cell inhibition ratio was accounted and the results formulated in a table. After 24 h, flow cytometric analysis was used to detect the cell cycle of the colonic carcinoma cell lines LoVo. Results Both proglumide and NS-398 inhibited the proliferation of LoVo cells and the combination of two reagents group synergistically inhibited the proliferation of LoVo cells, which bad a time dependent antiproliferation effect. FCM indicated that proglumide and NS-398 could both inhibit the growth of LoVo cells. Combination of the two reagents produced more significant inhibitive effects. Conclusions Both proglumide and NS-398 can inhibit the transition of Gx to S phase in the LoVo ceils, inhibiting them from entering the S phase, thus, repressing the cell proliferation. The combination can synergistically inhibit the proliferation of LoVo cells.
