摘 要:目的采用基因芯片技术比较筛选前后的人胃癌细胞母系MKN-28和亚系MKN-28S之间的基因表达谱,利用生物信息学方法筛选出与胃癌侵袭和转移相关的差异基因。方法用肿瘤转移基因芯片筛选出人胃癌细胞MKN-28母亚两系的转移相关基因,对部分有差异表达的基因用RT-PCR和Western blot进行分析鉴定。结果用肿瘤转移基因芯片筛选出两系的差异表达基因共20个,其中上调基因13个,下调基因7个。选择其中有代表意义的2个基因利用RT-PCR和Western blot进行验证,证实所选基因在筛选前后瘤细胞中均有差异表达,与基因芯片的表达情况一致,说明基因芯片结果可信度较高。结论基因芯片技术是筛选胃癌转移相关基因的有效方法;多基因参与了胃癌的侵袭和转移的过程,包括一些基因如Flt-4、SRC等基因和其它相关基因的改变。Objective To analyze the differences in gene expression between gastric cancer MKN -28 of parent and offspring cell lines using microarray technology and to screen gastric metastasis - associated genes by bioinformaties. Methods The metastasis - associated genes of parent and offspring cell lines were identificated using cDNA microarray of tumor metastasis - associated genes. The mieroarray results were confirmed by RT - PCR and Western - blot. Results The gene chip analysis showed that 20 genes were screened out for differences between the two generations solid tumor, which was involved in several aspects of tumor metastasis. There were 7 genes down - regulated and 13 up - regulated. Two genes among them were further confirmed by RT - PCR and Western - blot spectively and the expression of them was coincided well with the gene chip results. Conclusion cDNA microarray technique might be a useful method in screening gastric cancer metastasisas - sociated genes ; many different genes are involved in the invasion and metastasis of gastric cancer like Fit - 4, SRC as well as other relative genes.
