摘　　要：目的观察Caveolin-1在氧化型低密度脂蛋白(ox-LDL)诱导的巨噬细胞荷脂过程中的表达变化,进而分析Caveolin-1在巨噬源性泡沫细胞形成过程中的作用。方法采用75 mg/L ox-LDL与RAW264.7细胞共同孵育不同时间,高效液相色谱法检测细胞内胆固醇含量的变化,建立巨噬细胞荷脂化模型;Western-blot检测Caveolin-1蛋白的表达改变。结果75 mg/L ox-LDL与RAW264.7细胞共同孵育48 h后,细胞内胆固醇酯/总胆固醇的比值上升至42.3%±5.9%,符合荷脂细胞特征。Caveolin-1的蛋白表达较0 h组明显减弱。结论ox-LDL诱导的RAW264.7巨噬细胞荷脂化过程中,Caveolin-1的表达下调。Objective To observe the protein expression of Caveolin - 1 in lipid - loaded macrophages induced by ox-idized low density lipoprotein and explore the effect of Caveolin - 1 on the process. Methods RAW264.7 mouse macrophages were treated with 75 mg/L oxidized low density lipoprotein for different time（0 h, 24 h,48 hand 72 h ）. High performance liquid chromatography （ HPLC ） analysis was peformed to determine the content of cellular cholesterol. Western - blot was used to detect the protein expression of Caveolin-1. Results Western - blot and immunofluoreseence showed that the protein expression of Caveolin-1 was significantly decreased in a time - dependent manner in maerophages. Conclusion The protein expression of Caveolin - 1 was down - regulated in lipid - loaded macrophages.
