摘 要:目的探讨蛋白酶体抑制剂MG-132对体外人U87胶质瘤细胞增殖和凋亡的影响。方法分别以不同浓度(0、2.5、5、10和20μmol/L)的MG-132培养U87胶质瘤细胞,通过四甲基偶氮唑蓝(MTT)法检测不同培养时间(12、24、48、72 h)对U87胶质瘤细胞增殖的影响,流式细胞仪分析MG-132对U87胶质瘤细胞凋亡的影响,HE染色观察细胞形态学变化。结果通过与正常对照组比较,2.5-20μmol/L浓度的MG-132在12、24、48及72 h均可抑制U87细胞增殖并诱导其凋亡,并在一定范围内呈时间和剂量依赖性;各亚组与对照组差异具有统计学意义(P〈0.05);形态学检查呈现凋亡细胞的特征。结论MG-132可抑制U87胶质瘤细胞增殖并诱导其凋亡,在一定范围内呈时间和剂量依赖性。Objective To explore the effects of proteasome inhibitor MG-132 on proliferation, apoptosis of U87 glioma cells in vitro. Methods U87 glioma cells were cultured with different concentrations of proteasome inhibitor MG-132 (0,2.5,5,10 and 20 μmol/L) . Cell viability was determined by MTT assay at different cultured periods( 12,24,48,72) ,apoptosis rates were measured by flow cytometry (FCM), and HE were used to detect the morphological changes of apoptotic cells. Results Compared with normal control, exposure to MG-132 had a antiproliferative effect on U87 glioma cells and induced U87 glioma cell apoptosis, and was in a certain range of time and dose-dependence; In each sub-group and control group, differences had statistical significance(P 〈 0.05). Conclusion Proteasome inhibitor MG-132 can inhibit U87 glioma cell proliferation and induce U87 glioma cell apoptosis,in a certain range time and dose-dependent manner.
