非小细胞肺癌患者癌组织与外周血淋巴细胞p53蛋白表达相关性研究

摘　　要：目的探讨p53蛋白在非小细胞肺癌(NSCLC)患者癌组织和外周血淋巴细胞(PBL)中的表达及其相关性,以期建立更为实用简便的化疗敏感性预测指标。方法采用免疫荧光化学染色法检测49例手术NSCLC患者癌组织和PBL中突变p53蛋白的表达,并以健康人PBL中突变p53蛋白表达作为阴性对照。结果p53蛋白在癌组织、NSCLC患者PBL和健康人PBL中的表达分别为63.26%(31/49)、59.18%(29/49)和5%(1/20),其中癌组织的p53蛋白表达与NSCLC患者PBL的表达呈正相关,20例健康人PBL中p53蛋白表达阳性率显著低于NSCLC患者(P<0.01)。p53的表达在NSCLC不同临床病理类型及分期间差异无统计学意义(P>0.05),而在有无淋巴结转移患者间差异有统计学意义(P<0.01)。结论检测NSCLC患者PBL中p53蛋白表达可间接反应癌组织的p53蛋白表达状况。Objective To explore the correlation of the expression of p53 between non-small cell lung cancer （NSCLC） cell and peripheral blood lymphocytes （PBL）. Methods Expression of p53 were detected with immunofluorescence stain,in specimens and PBL of 49 patients with NSCLC. PBL of 20 normal people were used as control. Results The positive expression rate of p53 was 63.26 %, 59.18 % and 5 % in carcinoma tissues, PBL of NSCLC and PBL of normal people, respectively. The expression rate of p53 in carcinoma tissues was higher than that in PBL of normal people （P〈0.01） ,and positively correlated with that in PBL of the patients. The expression of p53 in carcinoma tissues and PBL of NSCLC was related to lymph node metastasis,but not to histological grade,subtype and TNM stage （P〉0.05）. Conclusion The expression of p53 in PBL of NSCLC patients may indirectly represent the expression of p53 in carcinoma tissues.
