摘 要:目的探讨在1-甲基-4-苯基-1,2,3,6-四氢吡啶(1-methyl-4-phenvl-1,2,3,6-tetrahydropyridine,MPTP)致C57BL/6小鼠多巴胺能神经元损伤模型中β-catenin在腹侧中脑不同时间点的表达变化。方法采用腹腔注射MPTP制备C57BL/6小鼠帕金森病模型。40只雄性C57BL/6小鼠,随机分为8组(n=5):对照组、MPTP 0 d组、MPTP 1 d组、MPTP 2 d组、MPTP 4 d组、MPTP 7 d组、MPTP 14 d组、MPTP 21 d组。在末次注射MPTP后的相应时间点将动物处死,Western blot法检测腹侧中脑磷酸化β-catenin(phosphorylatedβ-catenin,p-β-catenin)以及总β-catenin蛋白的表达;免疫组化法检测腹侧中脑β-catenin蛋白阳性细胞表达。结果MPTP致伤显著增加了C57BL/6小鼠腹侧中脑总β-catenin蛋白的水平,然而p-β-catenin蛋白并没有明显的改变。免疫组化检测显示MPTP处理后第7天β-catenin蛋白在腹侧中脑各区域黑质网状部(SNr),黑质致密部(SNpc)以及被盖腹侧区(VTA)的表达增多并发生了核转移。结论MPTP所致腹侧中脑区β-catenin蛋白表达增加与帕金森病发生发展的病理机制有关。Objective To investigate the change of the expression of β-catenin in ventral midbrain of 1-methyl-4-phenvl-1,2,3,6-tetrahydropyridine(MPTP)-induced domaminergic neurons injury model of C57BL/6 mice.Methods The Parkinson disease(PD) model in C57BL/6 mice was established by intraperitoneal injection of MPTP.Forty male C57BL/6 mice were randomly divided into 8 groups equally: control group,MPTP 0 day group,MPTP one day group,MPTP 2 day group,MPTP 4 day group,MPTP 7 day group,MPTP 14 day group,and MPTP 21 day group.The mice in each group were sacrificed at the corresponding time point after last MPTP injection.Western blot was used to detect the expression of phosphorylated β-catenin(p-β-catenin) and total β-catenin in ventral midbrain.By using immunohistochemistry,the cells positive for the expression of β-catenin in ventral midbrain were observed.Results MPTP lesion obviously increased the expression of total β-catenin in ventral midbrain in C57BL/6 mice,but there was no significant change in p-β-catenin in the ventral midbrain.Immunohistochemistry revealed that at the 7th day after MPTP injection,total β-catenin was expressed intensively and transferred to nuclear in ventral midbrain regions including substantia nigra pars reticulata(SNr),substantia nigra pars compacta(SNpc),ventral tegmental area(VTA).Conclusion The MPTP-induced increase in the expression of β-catenin in ventral midbrain is associated with the pathomechanism of PD.
