吗啡在楔状核伤害感受调制中作用的研究

摘  要:目的:观察中脑楔状核(NCF)尾部给予外源性吗啡对延髓中缝大核(NRM)和中脑导水管周围灰质(PAG)内痛兴奋神经元(PEN)的痛放电影响。方法:用玻璃微电极细胞外记录方法,分别在NRM和PAG内记录PEN的单位放电。结果:(1)NCF内注入吗啡(3×10-2mol/L)能明显抑制PAG、NRM内PEN的痛放电频率;(2)NCF内微量注入纳洛酮(2.74×10-3mol/L)能增加PAG、NRM内PEN的痛放电频率;(3)NCF内预先注射纳洛酮(5.5×10-3mol/L)能部分阻断吗啡对PAG、NRM内PEN痛放电抑制效应。结论:NCF内某些神经元与抗伤害性感受有关;Objective:To explore the effect of microinjecting morphine of naloxone into nucleus cuneiformis(NCF) on the frequencies of evoked discharge of pain excitation neurons (PEN) in periaqueductal gray(PAG) or nucleus raphe magnus(NRM).Methods:The extracellular recording method was used to investigate the effect of the antinociceptive role of opioids in NCF.Results:(1)Intra NCF injection of morphine(3×10 -2 mol/L,n=8) could suppress the frequencies of evoked discharge of PEN in PAG or NRM ( P <0 001,0 01,0 05),the maximal inhibitory rate was (77 3±7 5)% and (78 4±7 0)% respectively;(2)Naloxone(2 74×10 -3 mol/L,n=8)injected into NCF could enhance the evoked discharges of PEN in PAG and NRM( P <0 01,0 05),the percentage of change in the frequence of evoked discharge increased by (82 9±4 1)% and (79 5±3 1)% respectively.(3)Both of the above mentioned inhibitory effects of morphine (n=8) could be antagonised by intra NCF preinjection of naloxone (5.5×10 -3 mol/L,n=8)( P <0 001)。Conclusion:Some neurons in NCF might participate in the process to modulate noxious information transmission in CNS.Endogenous opiate might be a neuromodulator or transmitter in the antinociception of NCF.Both of the inhibitions of morphine injected into NCF on the PEN in PAG or NRM may be mediated by opioid receptor.
