摘　　要：目的检测干细胞标志物ABCG2在胃癌患者癌组织、癌旁组织中的表达,探讨其与临床病理因素的相关性及作为胃癌诊断新的分子标志物的可能性。方法收集62例胃癌患者手术切除的癌组织和对应的癌旁组织,用RT-PCR检测ABCG2的mRNA表达,用western blot和免疫组织化学在蛋白质水平上验证胃癌和癌旁组织ABCG2的表达,分析ABCG2在胃癌组织的表达与临床病理参数的相关性。结果胃癌组织ABCG2 mRNA的阳性表达率为61.3%(38/62),癌旁组织为11.3%(7/62),差异有统计学意义(P<0.01);在蛋白质水平上胃癌组织ABCG2的表达与肿瘤分化状态相关,低、未分化组高于中、高分化组(P<0.05),但与年龄、性别、肿瘤大小、浸润深度、TNM分期和有无淋巴结转移无关。结论胃癌组织中ABCG2的表达与肿瘤分化状态相关,可望作为胃癌诊断的新的分子标志物。Objective To detect the expression of stem cell marker ABCG2 in paired gastric cancer （GC） and adjacent non-cancerous tissues to explore whether ABCG2 is correlated with the clinicopathological parameters of GC and whether ABCG2 may serve as a novel molecular marker for GC. Methods The expression of ABCG2 in the tumor and adjacent non-cancerous tissues from 62 GC patients was examined by reverse transcription-PCR （RT-PCR）. Western blot （WB） and immunohistochemistry （IHC） were employed to detect ABCG2 protein. Results RT-PCR results showed that the positive rate of ABCG2 expression in the tumor tissues was 61.2% （38/62）, which was significantly higher than that in the adjacent non-cancerous tissues （P 〈 0. 01 ）. ABCG2 protein, verified by WB and IHC,in GC tissues was correlated with the differentiation, but not with the age, gender, tumor size, TNM stage, invasion depth, or the presence of lymph node metastasis. Conclusions ABCG2 expression in GC tissues is associated with the differentiation status. It may be a novel diagnostic marker for GC.
