摘 要:目的建立款冬花中款冬酮的超临界CO2萃取方法并与溶剂提取方法比较。方法采用超临界CO2萃取、溶剂提取法提取款冬花中款冬酮,并对提取物进行高效液相色谱分析。色谱条件为:色谱柱Diamonsil C18(200mm×4.6mm,5μm),流动相为甲醇∶水(77∶23),流速1ml/min,检测波长219nm。结果经均匀设计实验的筛选,得超临界CO2萃取优化的最佳条件为:萃取压力22MPa,萃取温度40℃,萃取时间35min。该条件下款冬酮得率为0.33%;常规溶剂提取法提取款冬酮的得率为0.17%。结论超临界CO2萃取方法能有效萃取款冬花中的款冬酮,款冬酮得率明显高于溶剂提取法,且纯度较高。该方法省时、易操作、提取效率高,可用于款冬花中款冬酮的提取。Objective To develop a method for extraction of tussilagone from Flos Farfarae using SFE-CO2 and to compare its effect with traditional solvent extraction. Methods The method of SFE-CO2 and solvent extraction was used to extract tussilagone from Flos Farfarae, and the uniform design was used to select SFE-CO2 extraction conditions. Quantification of the content of tussilagone in the extract was performed by HPLC on Diamonsil C 18 column (200 mm × 4.6 mm, 5 mm)using methanol-H20 (77:23 )as mobile phase and detection wavelength of 219 nm. Results The optimum extraction conditions for SFE-CO2 were as follows:pressure of abstraction:22 MPa;temperature of abstraction:40 ℃;time of abstraction:35 rain. The yield of tussilagone by SFE-CO2 was 0.33 % ,while the tussilagone yield by solvent extraction was 0. 17%. Conclusion SFE-CO2 is efficient in extracting the tussilagone from Flos Farfarae,and the yield is higher than traditional solvent extraction. The method is simple ,time-saving and efficient ,which can he used for extraction of tussilagone from Flos Farfarae.
