摘  要:目的:研究雌激素对绝经后雌性大鼠局灶性脑缺血-再灌注损伤后NF-κB基因表达和TNF-α含量的影响。方法:采用线栓法复制局灶性脑缺血-再灌注模型,应用RT-PCR法检测NF-κB基因的表达,利用放免法检测TNF-α含量。结果:与模型组大鼠比较,雌激素组脑缺血-再灌注后NF-κB表达明显减弱(P<0.01),TNF-α含量明显减少(P<0.01)。结论:雌激素能抑制大鼠脑缺血-再灌注后NF-κB表达,降低TNF-α含量,这可能是其脑保护作用的机制之一。Objective: To study the effects of estrogen on the expression of NF-KB and TNF-α in ischemia-reperfusion brain tissue of rats. Methods: Middle cerebral artery ischemia-reperfusion rats models were performed by modified fila- ment method. RT-PCR was used to detect the expression of NF-KB. Radioimmunity was used to examine the content of TNF-α. Results: Compared with the model group, the expression of NF-KB remarkably attenuated (P〈0.01), and the con- tent of TNF-α decreased significantly in the estrogen group (P〈0.01). Conclusion: Estrogen inhibits the expression of NF- KB, decreases the content of TNF-α after cerebral ischemia-reperfusion. This may be one of the mechanisms in which es- trogen performs its neuro-protection effect.
