丹皮药用组合物对酒精性脂肪肝大鼠的作用

摘　　要：目的:研究丹皮药用组合物对酒精性脂肪肝大鼠的作用及其机制。方法:采用56度二锅头酒灌胃SD大鼠复制酒精性脂肪肝模型,每日2次,共12周。第8周开始灌胃丹皮药用组合物,共4周。赖氏法测定血清丙氨酸转氨酶(alanine aminotransferase,ALT)和天冬氨酸转氨酶(aspartate aminotransferase,AST)的含量;4-氨基安替比林法检测肝脏和血清三酰甘油(triglyceride,TG)含量;酶比色法检测血清和肝脏游离脂肪酸(free fatty acids,FFA)含量;硫代巴比妥酸比色法检测血清和肝脏丙二醛(malondialdehyde,MDA)含量。结果:白酒灌胃8周后大鼠形成酒精性脂肪肝,经丹皮药用组合物治疗4周后,血清和肝脏TG水平、血清ALT和AST、血清和肝脏FFA及MDA均显著降低。结论:丹皮药用组合物具有抗大鼠酒精性脂肪肝作用,护肝、降脂及改善脂质过氧化状态可能是其主要作用途径。Objective. To study the protective effects of combination of medicinal components of Corteo： Moutan against alcoholic fatty liver （AFI.） and explore its mechanisms. Methods： The AFL model was induced by intragastric injection of Ergoutou wine in SD rats, twice daily during 12 weeks. The AFL rats were intragastrically administered with the combination of medicinal components of Corteoc Moutan on the 8th week, continuously for a total of 4 weeks. The serum levels of alanine aminotransferase （ALT） and aspartate aminotransferase （AST） were assayed using Reit＇s method. The contents of triglyceride （TG）, free fatty acids （FFA） and malondialdehyde （MDA） in the serum and liver were assayed using 4-aminoantipyrine coloring, enzymic colorimetry and thiobarbituric acid colorimetry, respectively. Results： Rat model of AFI. was induced successfully 8 weeks after intragastric injection of wine. In the groups treated with the combination of medicinal components of Corte3 Moutan, the levels of ALT, AST, TG. FFA and MDA in the serum and liver were significantly decreased 4 weeks after treatment. Conclusion： The combination of medicinal components of Corte~： Moutan possesses the effect against AFL in rats, and its main mechanisms may involve in the recovery of liver function, the decrement of lipids and the improvement of lipid peroxidation.
