摘  要:目的观察大蒜素对胃癌BGC823细胞生长以及对PPARγmRNA表达的影响,探讨大蒜素抑制BGC823细胞增殖的可能作用机制。方法不同浓度的大蒜素处理胃癌BGC823细胞,MTT法观察细胞增殖抑制作用;RT-PCR法检测胃癌BGC823细胞中PPARγmRNA表达的变化。结果大蒜素抑制BGC823细胞增殖,呈浓度和时间依赖性;大蒜素作用72h后明显抑制胃癌BGC823细胞PPARγmRNA表达,呈浓度依赖性。结论大蒜素可能通过下调PPARγmRNA表达抑制胃癌BGC823细胞增殖,发挥抗肿瘤作用。OBJECTIVE To investigate the effect of allicin on cell growth and expression of peroxisome proliferators-activated receptor gamma(PPARγ) in human gastric cancer cell line BGC823. METHODS BGC823 cells were treated with various concentration of alliein. Cell proliferation was assayed by MTT method and PPARTmRNA was semi-quantified by RT-PCR. RESULTS Allicin had a potent inhibitory effect on the growth in BGC823 cells in a dose-dependent and a time-dependent manner. After 72h treatment of allicin the expression of PPARγmRNA in BGC823 cells was significantly down-regulated. CONCLUSION Alliein can inhibit the growth of BGC823 cells which is associated with a down- regulation of PPARγmRNA.
