摘  要:目的优选超声波辅助热回流法提取马鞭石斛多糖工艺。方法以多糖的提取率作为指标,通过单因素及正交实验对提取工艺进行优化,主要考察因素为提取时间、温度和次数。结果超声波辅助热回流提取马鞭石斛多糖的最优工艺条件:温度95℃,时间16 m in,提取4次,采用此工艺提取的马鞭石斛多糖提取率为(18.64±0.396)%,显著高于传统热回流法的提取率。结论该工艺具有良好的精密度和稳定性,重复性好,适用于马鞭石斛多糖的提取。Objective To optimize the extraction process of polysaccharide from D.fimbriatum Hook.var.oculatum Hook by ultrasonic hot reflux assisted.Methods On the basis of single factor experiments,the extraction process was optimized by orthogonal experimental design on effects of extraction temperature,ultrasonic-irradiation time and extraction times.The yield of polysaccharide was used to evaluate the factor levels.Results The optimum conditions were determined as follows: ultrasonic-irradiation 16 min at 95℃ and extraction four times.The extraction rate of polysaccharide reached(18.64±0.40)% by using the method,which was much better than hot reflux extraction method.Conclusion That the method is simple with high accuracy,good stability and reproducibility,and can be used to extract polysaccharide from D.fimbriatum Hook.var.oculatum Hook.
