摘 要:探索具有抗肿瘤特性的中性粒细胞的筛选技术,为临床肿瘤的细胞治疗提供一种新策略。采用密度梯度离心法对5名健康者和2例肿瘤患者全血粒细胞进行分离,与肿瘤细胞进行混合,利用MTT和台盼兰计数法检测中性粒细胞增殖活性,用全自动化学发光仪测定中性粒细胞功能。结果表明:噻唑蓝(MTT)和台盼兰方法检测肿瘤细胞增殖活性均显示,2号受试者肿瘤细胞数最低,中性粒细胞化学发光结果显示,其中性粒细胞发光强度较强,提示2号受试者中性粒细胞杀伤肿瘤细胞能力较强。MTT和台盼兰方法具有很好的一致性,可作为具有抗肿瘤特性的中性粒细胞筛选的备选方法。To explore an effective method to choose polymorphonuelear leukocytes with anti-tumor trait. The neu- trophil from peripheral blood of 5 health people and 2 tumor patients were isolated by neutrophil separation me- dium and mixed with tumor cells. The function of neutrophils was detected by chemical luminescence method. The cell proliferation was observed by MTY and typran method. The results showed that the number of tumor cells for No. 2 sample was lowest among others and the neutrophils function of No. 2 sample was stronger than others. The MIT and typran method could be an effective method for choosing polymorphonuclear leukocytes with anti-tumor trait.
