表柔比星对乳腺癌细胞c-FLIP表达的影响

摘　　要：目的:探讨表柔比星对乳腺癌细胞中c-FLIP表达的影响。方法:以MCF-7及MDA-MB-231乳腺癌细胞株为对象分两组:处理组中加入2 mg/L的表柔比星分别作用24 h、48 h和72 h,对照组加入等量生理盐水。采用RT-PCR技术检测两组细胞株中c-FLIP的表达,流式细胞仪检测细胞凋亡百分率。结果:MCF-7及MDA-MB-231乳腺癌细胞中有c-FLIP的表达;与对照组相比,表柔比星作用24 h,乳腺癌细胞中c-FLIP的表达明显下降,48 h时c-FLIP表达有所升高,72 h时表达最低。随着表柔比星作用时间延长,细胞凋亡率逐渐增加,72 h达到最高。结论:表柔比星通过抑制c-FLIP的表达促进乳腺癌细胞的凋亡。Objective：To discuss the effect of epirubicin on the expression of c FLIP in breast cancer. Method： MCF-7 and MDA-MB-231 breast cancer cells incubated were divided into two group,epirubicin group were treated with epirubicin,and control group were treated with 0.9% sodium chloride solution at the same dose. After 24 h,48 h and 72 h,incubated cells were collected for the measurement of c-FLIP by RT PCR,and for examination of percent of apoptosis ceils with flow cytometry. Results ：There was expression of c-FLIP in MCF-7 and MDA-M-231 breast cancer cell lines. Compared to control group,c-FLIP expression was down regu Lated significantly after cell lines were treated 24h with epirubicin,c-FLIP expression rose softly at 48 b and c-FLIP reached the lowest level expression at 72 h. Percentage of cell apoptosis ascended gradually,along with time epirubicin functioning prolonged. It was the most grievous of cell apoptosis at 72 h in epirubicin group. Conclasion：Epirubicin can promote apoptosis of breast cancer cells by restraining the expression of c-FLIP.
