摘 要:目的:探讨Smad4基因在大肠癌中的表达及其与肿瘤分化程度或远处转移的关系。方法:利用免疫组织化学S-P法检测52例大肠癌组织、19例相应的癌旁正常组织中Smad4的表达。结果:Smad4在大肠癌原发灶中的阳性表达率为63.5%(33/52),与癌旁正常组织中的阳性表达率89.5%(17/19)相比,Smad4阳性表达水平显著降低,差别有显著性(P<0.05);Smad4与肿瘤细胞分化程度无关(P>0.05),但与淋巴结转移显著相关(P<0.05)。结论:Smad4的低表达可能是大肠癌发生过程的晚期事件,并可能通过直接或间接的作用促进大肠癌的淋巴结转移。Objective:To study the expression of Smad4 gene in colorectal carcinoma as well as its association with degree of differentationg and metastasis. Methods:The expression of Smad4 was detected in 52 cases with primary colorectal carcinoma and 19 adjacent normal colorecta tissues by using immunohistochemistrical S -P method. Results:Positive expression rate of Smad4 in primary colorecta cancer tissues was 63.5% (33/52), compared with the positive rate 89.5% (17/19) in adjacent normal colorec- ta tissues, the Smad4 expression level apparently degraded and there was significant differentiation ( P 〈 0.05 ). The positive ex- pression of Smad4 had no correlation with degree of differentationg( P 〉 0.05 ) ,but it was closely associated with lymph node metastasis ( P 〈 0.05). Conclusion: The low expression of Smad4 could be an advanced stage during the course of the colorectal carcinoma formation. Simultaneously, it also promoted lymph node metastasis directly or indirectly.
