摘　　要：目的：探讨人类疱疹病毒６型（ＨＨＶ６）与血液病患者巨噬细胞集落刺激因子（ＭＣＳＦ）异常表达的关系。方法：以卵白素生物素复合物（ＡＢＣ）免疫酶标和逆转录聚合酶链反应（ＲＴＰＣＲ）方法检测３０例血液病患者骨髓单个核细胞体外感染ＨＨＶ６后ＭＣＳＦ和ＭＣＳＦｍＲＮＡ的表达。结果：ＨＨＶ６感染组ＭＣＳＦ阳性细胞百分率、积分值和ＭＣＳＦｍＲＮＡ检出率分别为３６．９％±２４．９％，４５．９±３１．４和８３３％，显著高于对照组（２０．３％±１９．４％，２５．３±２４．２，５０．０％）（Ｐ＜０．００１）；其中急性白血病组和慢性粒细胞白血病组ＭＣＳＦ表达升高幅度较大。ＭＣＳＦ主要分布于粒细胞和单核细胞的胞膜和胞浆。Objective: To study the relationship between HHV 6 infection and abnormal expression of M CSF in bone marrow mononucleated cells (MNC) from patients with hematological disorders.Methods:M CSF expression level were detected in 30 specimens of MNC from patients with hematological diseases by ABC immunocytochemical staining and reverse transcription polymerase chain reaction methods.Results:The percentage and score of M CSF antigen positive cells, and percentage of M CSF mRNA expressing samples derived from HHV 6 infected group were 36.9%±24.9%, 45.9±31.4 and 83.3%, respectively,which were significantly higher than those of control (20.3%±19.4%,25.3±24.2 and 50.0%,respectively)(P<0.001).HHV 6 could promote M CSF production to a large extent in MNC from leukemic patients than that from patients with benign hematological disorders. M CSF existed mainly on membrane or in cytosol of granulocytes and monocytes.Conclusion:HHV 6 infection in vitro could promote M CSF production in MNC from patients with hematological diseases.
