摘 要:目的探讨同型半胱氨酸(Hcy)与谷氨酸单钠(MSG)联合作用建立的妊娠期高血压疾病脑病动物模型的发病机制。方法将孕鼠随机分为4组,每组10只。从孕10 d起,对照组(PN组)、妊娠谷氨酸组(PG组)均每日腹腔内注射生理盐水2 ml,同时分别背部皮下注射生理盐水及MSG[1 g(/kg.48 h)];妊娠同型半胱氨酸组(PH组)及妊娠同型半胱氨酸+谷氨酸组(PHG组)每日腹腔内注射Hcy 2 m[l200 mg(/kg.d)],同时分别背部皮下均注射生理盐水及MSG[1 g(/kg.48 h)];直至孕20 d。孕期测定大鼠血压、尿蛋白定量、行为改变及原位杂交法检测NMDAR-1 mRNA及GABA-A mRNA的表达。结果PH组及PHG组血压于12 d均明显上升,与PN组差异有显著性(P<0.01);PH组、PHG组与PN组相比,24 h尿蛋白定量均有明显增加(P<0.01);而PH组与PHG组相比,无明显差异;PG组与PN组相比,无明显差异;PHG组孕鼠出现阵发性凝视、一侧肢体震颤、面部抽搐等行为改变;PHG组脑组织各部位的NMDAR-1 mRNA的原位杂交阳性细胞表达与其他3组相比均有明显的增强(P<0.05);GABA-A mRNA表达的阳性细胞与其他3组相比均显著下降(P<0.05)。结论Hcy与MSG协同作用可引起孕鼠痫样发作,其发病机制可能与NMDAR-1 mRNA表达上调、GABA-A mRNA表达下调有关。Objective To explore the pathogenesis of eneephalopathy in the rat model of hypertensive disorder compheating pregnancy by injecting homocysteine (Hcy) and monosodium glutamate (MSG) in pregnant rats. Methods The pregnant rats were randomly divided into 4 groups (n =10 each group):group PN (control group,in which the rats were intraperitoneally injected with 2 ml of normal saline and subcutaneously injected with nomral saline from day 10 to day 20 of gestation ), group PC, (in which the rats were intraperitoneally injected with 2 ml of normal saline and subeutaneously injected with MSG from day 10 to day 20 of gestation),group PH (in which the rats were intraperitoneally injected with 2 ml of Hcy and subcutaneously injected with nomral saline from day 10 to day 20 of gestation), and group PHG (in which the rats were intraperitoneally injected with 2 ml of Hcy and subcutaneously injected with MSG from day 10 to day 20 of gestation). The blood pressure, urine protein, changes in behavior, and expressions of GABA-A and NMDAR-1 mRNAs were determined. Results Compared with groups PN and PG,the blood pressure significantly increased since day 12 of gestation in groups pH and PHG (P 〈 0.01 ). The amount of 24-hour urine protein was higher in groups PH and PHG than in groups PN and PG (P 〈 0.01 ). There was no significant difference between groups PH and PHG and between groups PG and PN. Obvious changes in the behavior were found in group PHG. Compared with other 3 groups,the expression of NMDAR-1 mRNA significantly inceased and the expression of GABA-A mRNA significantly decreased in group PHG (P 〈 0.05 ). Conclusion Hcy and MSG may act synergistically to induce eclampsia and preeclampsia, and the pathogenesis may be associated with the up-regnlated expression of NMDAR-1 mRNA and the down-regulated expression of GABA-A mRNA.
