摘  要:目的通过研究烟草悬凝物(CSC)对人支气管上皮细胞(BEP-2D cells)急性毒性损伤生物学作用,探讨烟草烟气在人支气管上皮细胞损伤过程中的机制,为呼吸道疾病的防治措施提供实验依据。方法收集某品牌香烟烟气并溶解于LHC-8培养液制备成CSC;采用永生化的人支气管上皮细胞,分为对照组(BEP-2D cells)和实验组(BEP-2DCSC cells)。观察烟草悬凝物染毒后的细胞存活率、膜通透性损伤、细胞凋亡率和次黄嘌呤磷酸核糖基转移酶基因(HPRT gene)突变率。结果在急性毒性损伤过程中,随着CSC浓度的逐渐增加:细胞存活率逐步下降、细胞培养液乳酸脱氢酶(LDH)逐渐且明显增多、细胞的早期和晚期凋亡率均增大、HPRT基因突变率逐渐增大,均与染毒剂量具有明显量-效关系。结论CSC作为一种复杂的香烟烟气混合物具有较为典型的急性细胞毒性作用,其机制可能与细胞膜通透性增加、细胞凋亡及HPRT基因突变有关。Objective To investigate the cytotoxicity, genotoxieity and its relevant mechanism of immortalized human bronchial epithelial ceIls(BEP-2D cells) treated by cigarette smoke condensate (CSC), and explore the chemical and biological effect during the damage of cells treated by CSC, and provide the new theoretical basis for diagnosis and treatment of lung cancer. Methods The smoke from the cigarette was collected by smoking machine and dissolved in serum-free cell culture medium LHC-8. Immortalized human bronchial epithelial cells were divided into two groups, named control group (BEP-2D cells) and experimental group (BEP-2D CSC cells ), for investigating the cytotoxicity, genotoxieity, oneogenic transformation and relative mechanism of CSC. Results With the concentration increased during the acute toxicity of BEP-2D cells treated by CSC, the treated cells had lower survival, more obvious cell membrane damage, and higher apoptosis rate that was in good correlation with the concentration of CSC during the early stage and late stage. Conclusion As a complicated commixture, the CSC has typical toxicity and its mechanism possibly concern the cell membrane permeability increases, the cell apeptosis and the HPRT gene mutation.
