乳腺癌中p-JNK蛋白的表达及其临床意义

摘　　要：目的研究磷酸化应激活化蛋白激酶(p-JNK)在乳腺癌癌变过程中的表达及其与临床病理特征的关系。方法采用免疫组织化学S-P法检测118例乳腺石蜡包埋组织中p-JNK蛋白的表达情况,采用Western blot检测30例乳腺浸润性导管癌及癌旁乳腺组织中p-JNK的表达情况。结果免疫组化结果显示,在乳腺癌癌变过程中p-JNK蛋白表达阳性率呈递减趋势;乳腺浸润性导管癌中p-JNK蛋白与患者年龄、肿瘤大小、淋巴结转移无相关性,与组织学分级和临床分期相关(P=0.015,P=0.012)。Western blot结果显示,乳腺浸润性导管癌中p-JNK蛋白的表达明显低于癌旁正常组织(P=0.014)。结论p-JNK蛋白在乳腺癌癌变过程中发挥重要的作用。Objective To investigate the expression and clinicopathological significance of p-JNK protein in the breast carcinogenesis. Methods The expression of p-JNK protein was examined in 118 paraffin-embeded specimens by immunohistocbemistry,including breast normal ductal hyperplasia, wildly and moderately atypical hyperplasia, severely atypical hyperplasia and intraductal carcinoma in situ, as well as invasive ductal carcinoma. Thirty cases of frozen breast cancer and pairing non-neoplastic tissue were subjected to Western blot for p-JNK protein. The relationship between the p-JNK expression and clinicopathological parameters of carcinomas was analyzed by Chi square test and t test. Results The positive rote of the p-JNK protein expression was gradually decreased in the breast carcinogenesis. No significant correlation was found between the p-JNK protein expression and age, tumor size or lymph node metastasis of carcinomas. However, the p-JNK protein expression was closely linked to histological grading and clinical stage （P = 0.015,P = 0.012 , respectively ）. Western blot analysis showed a significant decrease in the p-JNK protein expression in breast cancer tissues, compared with the adjacent normal breast tissues （P = 0.014）. Conclusion p-JNK protein plays an important role in breast carcinogenesis.
