摘 要:目的:观察中药龟鹿益髓胶囊(GLYS)对实验性脊髓空洞症模型兔脊髓神经元凋亡的影响。方法:48只中国白兔随机分为4组,经皮枕大池穿刺缓慢抽出0.6ml脑脊液,注入等量25%Kaolin制成脊髓空洞症模型。GLYS治疗组当日起给予不同剂量的龟鹿益髓混悬液灌胃,模型组给予等量生理盐水灌胃。核磁共核扫描检测空洞形成,并记录死亡率。取脊髓行HE、TUNEL及Bcl-2和Bax染色观察。结果:实验性脊髓空洞症兔出现颈项强直、抽搐、肢体瘫痪及运动功能障碍等症状。GLYS组动物后肢运动功能明显提高。模型组动物死亡率(5/12)明显高于龟鹿益髓组动物死亡率(2~3/12)(P〈0.05,或P〈0.01)。MRI显示有空洞形成,在同一时间点治疗组较模型组脊髓组织水肿减轻,空洞发生率较模型组低(P〈0.01)。TUNEL法检测神经元凋亡,治疗组均较少。治疗组Bax反应阳性细胞率低于模型组,Bcl-2反应阳性细胞率高于模型组。结论:中药龟鹿益髓胶囊明显减少实验性脊髓空洞症模型空洞形成,并明显减少脊髓神经元凋亡。Objective:To observe the effect of Guiluyisui(GLYS) capsule on the apoptosis of neurons in experimental syringomyeliain rabbit. Methods:48 Chinese white rabbits were divided randomly into four groups and 0.6ml cerebrospinal fluid were slowly draw from the cisteinamagna under ketamine anesthesis. The rabbits were subjected to injection of 25% kaolin mixed to produce the experimental syringomyeliain models. The rabbits in GLYS group were treated at the same time with different dose of GLYS by suspl intragastric administration and those in model were given the same volume normal saline by intragastric administration. The spinal cords were observed with hematoxylin and eosin ( HE ), TUNEL stain, Bcl-2 and Bax immunohistochemical method. Results: Experimental syringomyeliain rabbits taken on the neck rigidity, hyperspasmia temporarily, limbs palsy and motor dysfunction. The hind limb motor function of the experimental rabbits in GLYS were improved obviously. The mortality rate of model group was higher than GLYS groups ( P 〈 0.05, or P 〈 0.01 ). The porosis was observed in the spinal by MRI and the porosis rate of model group was much higher than GLYS groups. The hydrops of the myeloid tissue in treatment groups was better than that in model group at the same time point while the rate of porosis of treatment was lower than that of model group( P 〈 0.01). The rates of Bax positive cell in treatment groups were lower than that in model group while the rates of Bcl-2 in treatment groups were higher than that in model group. Conclusion : Guiluyisui capsule, traditional Chinese medicine agent, could obviously imporve the ischemic and hydrops of the spinal cord and reduce the spinal neuron apoptosis of experimental syringomyeliain rabbits.
